Pnömonili Koyun Akciğerlerinde Mycoplasma Türlerinin Moleküler Olarak Araştırılması

Öz

Bu çalışmada, Şanlıurfa mezbahanesinde kesilen ve makroskopik olarak pnömonik lezyonlar tespit edilen koyun akciğer örneklerinden Mycoplasma türlerinin varlığının moleküler tekniklerle araştırılması amaçlandı. Bu amaçla Siverek bölgesinde, 10 farklı sürüden 10’ar akciğer olacak şekilde toplam 100 adet akciğer numunesi alındı. Alınan akciğerler soğuk zincirde Harran Üniversitesi Veteriner Fakültesi Mikrobiyoloji Anabilim Dalı Laboratuvarına getirildi. Yapılan makroskobik incelemeler sonucunda pnömonik lezyonlar gözlemlenen örneklerden, DNA ekstraksiyon ve kültür-PCR yöntemi ile incelemek üzere iki seri örnek alındı. Doku örneklerinden ve aynı zamanda da ekim yapılan besi yerlerinden DNA izolasyonu gerçekleştirildi. Elde edilen DNA örnekleri, tür spesifik primer çiftleri kullanılarak M. ovipneumoniae, M. mycoides subsp. capri, M. capricolum subsp. capripneumoniae, M. capricolum subsp. capricolum, M. agalactiae ve M. arginini yönünden PCR yöntemi ile incelendi. Sadece kültür-PCR ile beş örnek (%5) M. mycoides subsp. capri yöününden pozitif olarak değerlendirilirken hem direkt hem de kültür-PCR tekniklerinde araştırılan diğer mikoplazma türleriyle ilişkili pozitif sonuç elde edilmedi. Çalışmada kültür-PCR tekniğinin etkeni saptamada dokudan direkt PCR tekniğine göre daha avantajlı olduğu görüldü. Sonuç olarak bölgedeki pnömoni vakalarında etiyolojik olarak M. mycoides subsp. capri’nin rolü olduğu kanısına varıldı.

Molecular Investigation of Mycoplasma Species in the Lungs of Sheep with Pneumonia

Öz

This study aimed to investigate the presence of Mycoplasma species in macroscopically detected pneumonia lesions in the lungs of sheep slaughtered in a slaughterhouse in Şanlıurfa using molecular techniques. For this purpose, a total of 100 lung samples were collected from ten different flocks in the Siverek region, each represented by ten lungs with macroscopic lesions. The samples were transported under cold chain conditions to the Laboratory of the Department of Microbiology, Faculty of Veterinary Medicine, Harran University. From each sample evaluated as pneumonic, two serial tissue portions were collected: one for DNA extraction from tissue to be used in direct PCR and the other for examination using the culture-PCR method. DNA was isolated from both tissue samples and cultured media. The DNA samples were tested using species-specific primer pairs targeting M. ovipneumoniae, M. mycoides subsp. capri, M. capricolum subsp. capripneumoniae, M. capricolum subsp. capricolum, M. agalactiae, and M. arginini. Five samples (5%) were found positive for M. mycoides subsp. capri by culture-PCR, while no positive results were obtained for other Mycoplasma species by either direct PCR or culture-PCR. The study revealed that culture-PCR was more advantageous than direct PCR for detecting the causative agent. In conclusion, M. mycoides subsp. capri was identified as an etiological factor in pneumonia cases in the region.
