Kodlama Yapmayan Uzun RNA’ların Mekanizması ve Klinik Yansımaları

Öz

Long non-coding RNA’lar (lncRNA’lar), 200 nükleotitten uzun ve protein kodlamayan RNA transkriptleridir; son yıllarda gen ekspresyonunun çok katmanlı düzenlenmesinde üstlendikleri rollerle dikkat çekmektedirler. Başlangıçta genomun "gürültülü" ürünleri olarak değerlendirilseler de, epigenetik modifikasyonlardan post-transkripsiyonel düzenlemeye kadar geniş bir etki yelpazesinde görev aldıkları gösterilmiştir. Yapısal çeşitlilikleri ve genetik konumlarına göre sınıflandırılan LncRNA’lar, moleküler etki mekanizmalarındaki fonksiyonel esneklikleriyle öne çıkmaktadır. Güncel çalışmalar, bu moleküllerin hücre proliferasyonu, farklılaşma, apoptozis ve bağışıklık regülasyonu gibi temel biyolojik süreçlerde önemli düzenleyiciler olduklarını ortaya koymaktadır. Bu işlevsel çeşitlilik, LncRNA’ların pek çok hastalıkta potansiyel biyobelirteç ve terapötik hedef olabileceğini göstermektedir. Bununla birlikte, LncRNA’ların evrimsel korunma eksikliği, düşük ekspresyon seviyeleri ve karmaşık ikincil yapıları, fonksiyonel karakterizasyonlarını zorlaştırmaktadır. Gelişen biyoteknolojik araçlar ve çoklu omik yaklaşımlar sayesinde bu sınırlılıkların aşılması beklenmekte ve LncRNA’lar, gelecek yıllarda hem temel bilim hem de klinik araştırmaların merkezinde yer alacaktır.

Mechanisms and clinical implications of long non-coding RNA’s

Öz

Long non-coding RNAs (LncRNAs) are RNA transcripts longer than 200 nucleotides that do not encode proteins, and in recent years they have attracted attention for their roles in the multilayered regulation of gene expression. Initially considered as “transcriptional noise” of the genome, they have since been shown to function across a wide spectrum of processes, from epigenetic modifications to post-transcriptional regulation. Classified according to their structural diversity and genomic location, LncRNAs stand out for their functional flexibility in molecular regulatory mechanisms. Recent studies have revealed that these molecules serve as key regulators in fundamental biological processes such as cell proliferation, differentiation, apoptosis, and immune regulation. This functional diversity highlights their potential as biomarkers and therapeutic targets in various diseases. However, their lack of evolutionary conservation, low expression levels, and complex secondary structures continue to pose challenges for functional characterization. With the advancement of biotechnological tools and multi-omics approaches, these limitations are expected to be overcome, and LncRNAs are anticipated to become central to both basic research and clinical investigations in the coming years.
