İNTEGRİN ΑIIBΒ3 (GPIIB-IIIA)’NIN AGREGASYONDAKİ ROLÜ

Öz

Tombositlerin aktivasyonu sonucunda trombositlerin birbirine eklenmesiyle trombosit agregasyonu gerçekleşir. ADP gibi agonistlerin trombosit ve hasarlı damar tarafından salınmasından sonra, trombositler şekil değişikliğine uğrar ve GPIIb-IIIa (αIIbβ3) reseptör bölgeleri açığa çıkar. Bu süreçteki ana trombosit reseptörü GPIIb/IIIa'dır. Fibrinojen köprüleri sayesinde aktif trombositler birbirlerine bağlanır. Trombositin uyarılması, granül egzositozu ile GPIIb/IIIa moleküllerinin yüzeye yerleşmesinde artışa ve GPIIb/IIIa aktivasyonuna yol açar. αIIbβ3’ün trombosit agregasyonuna aracılık etmedeki anahtar rolü ve önceki yıllarda bu rolün blokajının agregasyon yanıtını inhibe ettiğinin gösterilmiş olması bu reseptörü antitrombotik tedavi için hedef haline getirmiştir. αIIbβ3’ün hücre dışı parçasında oluşan yapısal değişiklikler, ligandlara ilgisini artırır. İntegrin reseptörlerindeki affinite düzenlenmesini içeren değişiklikler kendiliğinden gelişir. Trombositin agonist ile uyarılmasından sonra başlayan olaylar “iç-dış” sinyalizasyon olarak değerlendirilir.  αIIbβ3 ile tetiklenen sinyaller bir dizi moleküler ve hücresel olayları başlatır. Ligand αIIbβ3‘ten çıkan sinyaller “dış-iç” sinyalizasyon olarak tanımlanır. Primer gevşek agregasyon iç-dış sinyalizasyona bağımlı iken, sekonder sıkı agregasyon “dış-iç” sinyalizasyona bağımlıdır.
