Endemik Teucrium leucophyllum (Lamiaceae) tohumlarında in vitro çimlendirme çalışmaları

Öz

Teucrium leucophyllum Montbret & Aucher ex Bentham (Lamiaceae), Erzincan yöresinde (Türkiye) dar yayılış alanına sahip nadir endemik bitki türlerindendir. Bu çalışmada Teucrium leucophyllum türünde farklı NaCl, HCl, KNO3, GA3 konsantrasyonları, sıcak-soğuk stratifikasyon ve mekanik skarifikasyon uygulamalarının çimlenme başarısına etkilerinin incelenmesi amaçlanmıştır. Tohumlar 100 ve 200 μM konsantrasyonlarda NaCl, HCl, KNO3, GA3 solusyonları kullanılarak ya da sıcak-soğuk ön işlem ve mekanik uygulamalarının ardından 23/18°C’de 8 saat aydınlık/16 saat karanlık ortamda çimlendirilmiştir. Kontrol grubuyla karşılaştırıldığında (% 13.5), çimlenmenin 100-200µM GA3 (% 52.5 ve % 51.5), için 100-200 µM KNO3 (% 29 ve %28), zımpara ve 4°C’de ön üşütme uygulamalarıyla %25 oranında arttığı (DMRT, P0.05) gözlenmiştir. Çimlendirme hızı katsayısının ise en yüksek giberellik asit uygulamalarında (11.4 ve 13.09) olduğu bulunmuştur. Bu çalışma sonucunda, elde edilen veriler türün ex-situ korunması amacıyla kullanılabilecektir.
