THE MOLECULAR ROLE OF THE WAVE REGULATORY COMPLEX IN CANCER METASTASIS: A COMPREHENSIVE OVERVIEW

Cancer metastasis is among the most critical challenges in oncology and accounts for a major share of cancer-related deaths. This progression necessitates the active motility of cancer cells, a phenomenon closely tied to the dynamic remodeling of the actin cytoskeleton. This review focuses on the molecular composition, regulatory dynamics, and diverse roles of the WAVE Regulatory Complex (WRC)—a pivotal player in cell migration—within the context of cancer dissemination, drawing upon the latest literature. Structurally, the WRC comprises five subunits that transmit signals from small GTPases such as Rac1 to the Arp2/3 complex, initiating actin polymerization. This process facilitates the formation of invasive cellular extensions like lamellipodia, which are instrumental in migratory behavior. Accumulating evidence reveals that WRC components are highly expressed in various aggressive malignancies, correlating strongly with enhanced invasion potential, increased metastatic capacity, and poor clinical outcomes. Thus, WRC-mediated signaling pathways emerge as promising therapeutic targets, potentially contributing to the design of novel anti-metastatic strategies aimed at halting cancer spread.
