In vitro Koşullarda Humik Maddelerin Kolza Bitkisinin Fizyolojik Özellikleri, Sürgün Rejenerasyonu ve Antioksidant Enzim Aktiviteleri Üzerine Etkisi

Abstract

Bu çalışmada, hem katkı maddesi hem de bitki büyüme düzenleyici olarak seçilen farklı konsantrasyonlardaki potasyum humat, bor
humat ve demir humat'ın kolzanın fizyolojik özellikleri, sürgün rejenerasyonu ve antioksidant enzim aktiviteleri üzerine etkisi araştırılmıştır.
Steril edilen tohumlar % 3 (w/v) sükroz ve % 0.7 (w/v)agar ile herbiri 1 ppm, 3 ppm, 5 ppm and 7 ppm konsantrasyonlarında olan potasyum
humat, bor humat ve demir humat içeren Murashige and Skoog temel besiyerinde kültüre alınmıştır. Fizyolojik özellikler ve antioksidant
enzim aktiviteleri bir haftalık bitkicikler üzerinde gözlenmiştir. 3-4 haftalık bitkiciklerden alınan gövde eksplantları 1 mgl-1
6-
benzylaminopurine ve 0,2 mgl-1 1-naphthaleneacetic acid içeren besiyerinde kültüre alınmıştır. Çalışmanın sonuçları incelendiğinde, bütün
humik madde formlarının kolzanın fizyolojik özellikleri üzerinde olumlu etkisi olduğu, antioksidant enz,im aktivitesi üzerinde negatif etksi
olduğu ve sürgün rejenerasyonuna hiç bir etkisi olmadığı görülmüştür.

The Effects of Humic Substances on Physiological Properties, Shoot Regeneration and Antioxidant Enzymatic Activities in vitro Cultured Rapeseed

Abstract

It was investigated the effects of various concentrations of potassium humate, boron humate and iron humate which was selected the additives to basal medium and phytohormones on physiological characteristics, antioxidant enzymatic activities and shoot regeneration rapeseed in vitro. Sterilized seeds were cultured on Murashige and Skoog Basal Medium with 3% (w/v) sucrose and 0.7% (w/v) agar containing each of the individual 1 ppm, 3 ppm, 5 ppm and 7 ppm potassium humate, boron humate and iron humate. Physiological characteristics and antioxidant enzymatic activities were observed from one week old seedlings obtained from the seeds. For observing shoot regeneration capacity, stem segments 3-4 mm in length were excised from 3-4 week old seedlings obtained from the seeds. Stem explants were cultured on Murashige and Skoog Basal Medium with 1 mgl-1 6-benzylaminopurine and 0,2 mgl-1 1-naphthaleneacetic acid. As a results, it was expressed that all the humate forms had positive effects on physiological parameters, but there were no effect on shoot regeneration. It was observed negative correlation between antioxidant enzymatic activities and increasing doses of all the humate forms
