Effects of Erdostein Use in Pregnancy on Fetal Rat Lung Maturation

Abstract

Objective: This study was planned for investigating the probable effects of 2 and 4 mg erdostein use on fetal lung maturation Materials and Methods: Wistar-Albino pregnant rats were divided into 3 groups as group I (n=10): SHAM; group II (n=10): Erdosteine 2mg, group III (n=10):Erdosteine 4 mg. The medication were given orally to pregnant rats at the end of first week and continued until the day of fetus excision by laparotomy at day 13 or day 18. Following the delivery, birth weight, crown-rump length, weight of whole lung were measured. The maturation of lungs were evaluated histopathologically. Fetuses were grouped according to the developmental stages of lungs. Results : There were no significant differences in birth weight and crown-rump length between the group I, II and III for the fetuses of day 13. Only whole lung weights of group III was significantly higher compared to group I (p<0.05). Whole lung weights and crown-rump length of group III and group II at day 18 were statistically higher than group I. On gestation day 18, canalicular+saccular stage in fetal rat lung in group III (%50) was significantly higher than the Group I (%10) and Group II (%15). Conclusion: High doses and long-term use of erdosteine in pregnants rats has created a significant increase in the lung maturation of preterm excised fetus. Gebelikte Erdostein Kullanımının Sıçan Fetus Akciğer Matürasyonu Üzerine Olan Etkileri

Öz

Amaç: Bu çalışma 2 ve 4 mg erdostein kullanımının fetusların akciğer olgunlaşması üzerine olası etkilerinin araştırılması için planlandı. Gereç ve Yöntem: Wistar albino cinsi gebe sıçanlar: grup I (n=10): SHAM; grup II (n=10): 2 mg erdostein, grup III (n=10): 4 mg erdostein olarak 3 gruba ayrıldı. Gebe sıçanlara gebeliğin birinci haftası sonunda ilaç uygulaması başlandı ve sıçan fetuslarının 13. ve 18. günlerde laparotomi ile alındığı günün öncesine kadar devam edildi. Laparotomiyi takiben batın dışına alınan fetusların fetal doğum ağırlıkları, baş-makat uzunlukları, tüm akciğer ağırlıkları ölçüldü. Histopatolojik olarak akciğer matürasyonu değerlendirildi. Fetuslar akciğer gelişim evrelerine göre gruplandırıldı. Bulgular: Grup I , II ve III arasında 13 günlük fetuslarda doğum kilosu, baş-makat uzunluğu açısından anlamlı fark yoktu. Sadece akciğer ağırlığı açısından grup III değerleri grup I' e göre anlamlı olarak yüksekti (p<0.05). 18 günlük sıçanlarda ise fetal akciğer ağırlıkları ve baş-makat uzunluğu açısından grup III ve II değerleri grup I' e göre anlamlı derecede yüksek bulundu (p<0.05). 18 günlük fetal sıçan akciğerinde grup III de kanaliküler+sakküler evre oranı (%50), grup I (%10) ve grup II (%15) ye göre anlamlı olarak fazlaydı( p<0.05). Sonuç: Gebe sıçanlarda uzun süreli ve yüksek doz erdostein kullanımı preterm olarak alınan fetusların akciğer maturasyonunda anlamlı artış yaratmıştır.
