İn Vitro Fertilizasyon ve İntrastoplazmik Sperm Enjeksiyonu İçin Hyaluronik Asit'e Bağlanma İle Sperm Seçme Tekniği

Öz

Abatract: Quality of sperm used for in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) increases success rate of good quality embryo production. Sperm selected only on the basis of morphology by ICSI operator leads to embryo development with anormal chromosome, high rate of abort and serious develop- ment risk. New method evaluating maturation and function of sperm with the sperm bind to hyaluronic acid (HA), was developed. Sperms binding to HA have normal morphology, low rate of aneuplody and apoptosis, and are live and mature. Fertilization, implantation and rate of pregnancy and good embryo quality are increasing with sperm selected with HA. Binding sperm to HA can be used for routine semen analysis, IVF and ICSI in clinical diagnosis of male infertility. In conclusion, IVF and ICSI techniques can be improved with sperms se- lected by binding to HA and which is mature and has DNA integrity

Öz

İn vitro fertilizasyon (IVF) ve intrasitoplazmik sperm enjeksiyonu (ICSI) için kullanılan spermin kalitesi, bu teknikler ile kaliteli embriyo üretiminin başarı oranını büyük oranda arttırmaktadır. ICSI operatörü tarafından sadece morfolojiye dayalı seçilen sperm, anormal kromozomlu embriyo gelişimine, yüksek oranda aborta ve ciddi gelişim riskine yol açabilmektedir. Spermin hyaluronik asit’e (HA) bağlanması ile sperm olgunluğunu ve fonksiyonunu değerlendiren yeni bir teknik geliştirilmiştir. İn vitro olarak hyaluronik aside bağlanan spermler, düşük anöploidi ve apoptozis gösteren, normal morfolojiye sahip canlı ve olgun spermlerdir. HA ile seçilen sperm ile fertilizasyon, implantasyon, gebelik oranı ve embriyo kalitesi artmaktadır. Spermin HA’ya bağlanması, erkek infertilitesinin klinik tanısında, rutin semen analizi, IVF ve ICSI için yararlı bir şekilde kullanılabilmektedir. Sonuçta üstün niteliklere sahip ırklarda HA’ya bağlanarak seçilen olgun ve DNA bütünlüğüne sahip spermler ile IVF ve ICSI yöntemleri geliştirilebilir ve kaliteli embriyo üretilebilir.
