Topraktan izole edilen aktinomisetlerin lipaz aktivitelerinin belirlenmesi

Abstract

Bu çalışmada farklı toprak örneklerinden izole edilmiş 156 aktinomiset izolatı kullanılmıştır. İzolatlar uygun besiyerine ekilmiş ve 28°C’de 7-10 gün inkübe edilmiştir. İzolatların kültürel ortamda lipolitik aktivitelerinin incelenmesi amacıyla, hazırlanan lipaz besiyerinde izolatların oluşturdukları şeffaf zon cetvel ile ölçülmüştür. Bunlardan 53 tanesi (%34) 10 mm ve daha az, 99 tanesi (%63) 20 mm ve daha az, 4 tanesi (%3) ise 20 mm’den daha fazla şeffaf zon oluşturmuştur. Tribütirinli besiyerinde en fazla şeffaf zon oluşturan 47 numaralı (21,5 mm) izolatın 16S rRNA gen bölgesine göre yapılan dizi analizi sonucunda GenBank numarası EU283368.1 olan ‘uncultured actinobacterium clone AS42 16S ribosomal RNA gene’ e %97 oranında benzerlik gösterdiği görülmüştür. İzolatın elde edilen 16S rRNA dizi bilgisi GenBank’a KY172825 erişim numarası ile kaydedilmiştir.

Determination of lipase activities of actinomycetes isolates from soil

Abstract

In this study 156 actinomycetes isolates which were isolated from different soil samples were used. Isolates were inoculated proper medium and incubated at 28°C for 7-10 days. In order to investigate the lipolytic activities of the isolates in the culture medium, the transparent zone formed by isolates in the prepared lipase medium was measured with a ruler. 53 of them (34 %) 10 mm and less, 99 of them (63 %) 20 mm and less, 4 of them (3 %) more than 20 mm transparent zone produced. The isolate which showed the highest lipase activity molecular identification has been made which based on 16S rRNA gene sequence. The 16S rRNA sequence information obtained from the isolate was recorded to GenBank with the access number KY172825. As a result of the study has been made on the NCBI database by using BLAST program, it was observed that, isolate showed 97% similarity ‘uncultured actinobacterium clone AS42 16S ribosomal RNA gene’ which’s access number on the database is EU283368.1.
