Production of virus eliminated seed potato material by meristem culture in certain potato varieties cultivated in the Aegean Region

Abstract

For the present study, 321 tuber samples from 17 potato cultivars grown in Aegian region were collected and they were screened by DAS-ELISA for the presence of viruses named PVX, PVY, PLRV, PVS and PVM. The results showed that 14.64 % of tuber samples were infected by PVX, PVY, PLRV, PVS, PVS+PLRV, PVS+PLRV+PVY, PVS+PLRV+PVY+PVM. After breaking dormancy of tubers, 175 meristem were isolated from 47 tuber samples of 14 potato cultivars. 120 plantlets developed from meristems of Agria, Granola, Hermes, Latona, Marabel, Marfona, Milva, Russet Burbank, Resy, Sante and Shepody cultivars. The results revealed that 83.33 % of them were virus-free. PVY infection was determined in plantlets of Latona and Marfona cultivars, while PLRV was found in plantlets of Sante cultivar. In the mentioned cultivars, the elimination rates for viruses in question were 69.56 %, 68.42 % and 70.83 % respectively. When 444 virus-free plantlets which are multiplicated by node culture transferred to soil, 223 of plantlets (50.22 %) from 9 cultivars in the exception of Marabel and Milva survived and 143 (64.12 %) of them showed the formation of mini tubers. A total of 462 mini tubers were obtained in varying quantities according to varieties and the DAS-ELISA test results revealed that they were free of PVX, PVY, PLRV, PVS and PVM.
