Production of virus eliminated seed potato material by meristem culture in certain potato varieties cultivated in the Aegean Region

Abstract

For the present study, 321 tuber samples from 17 potato cultivars grown in Aegian region were collected and they were screened by DAS-ELISA for the presence of viruses named PVX, PVY, PLRV, PVS and PVM. The results showed that 14.64 % of tuber samples were infected by PVX, PVY, PLRV, PVS, PVS+PLRV, PVS+PLRV+PVY, PVS+PLRV+PVY+PVM. After breaking dormancy of tubers, 175 meristem were isolated from 47 tuber samples of 14 potato cultivars. 120 plantlets developed from meristems of Agria, Granola, Hermes, Latona, Marabel, Marfona, Milva, Russet Burbank, Resy, Sante and Shepody cultivars. The results revealed that 83.33 % of them were virus-free. PVY infection was determined in plantlets of Latona and Marfona cultivars, while PLRV was found in plantlets of Sante cultivar. In the mentioned cultivars, the elimination rates for viruses in question were 69.56 %, 68.42 % and 70.83 % respectively. When 444 virus-free plantlets which are multiplicated by node culture transferred to soil, 223 of plantlets (50.22 %) from 9 cultivars in the exception of Marabel and Milva survived and 143 (64.12 %) of them showed the formation of mini tubers. A total of 462 mini tubers were obtained in varying quantities according to varieties and the DAS-ELISA test results revealed that they were free of PVX, PVY, PLRV, PVS and PVM.

Ege Bölgesi’nde yetiştirilen bazı patates (Solanum tuberosum L.) çeşitlerinde meristem kültürü yöntemi ile virüslerden arındırılmış üretim materyali elde edilmesi

Abstract

Bu araştırmada Ege Bölgesi'nde yetiştirilen 17 patates çeşidine ait 321 yumru örneği PVY, PLRV, PVX, PVS ve PVM adlı virüslerin varlığı yönünden DAS-ELISA yöntemi ile testlenmiştir. Toplanan yumru örneklerinin % 14.64 oranında PVX, PLRV, PVY, PVS, PVS+PLRV, PVS+PLRV+PVY, PVS+PLRV+PVY+PVM virüs(ler) ile enfekteli olduğu tespit edilmiştir. Virüs ile enfekteli olan 14 patates çeşidine ait 46 adet yumru örneğinden dormansi periyodu sonrası 175 adet meristem izole edilmiştir. Agria, Granola, Hermes, Latona, Marabel, Marfona, Milva, Russet Burbank, Resy, Sante ve Shepody çeşitlerine ait 120 adet fide gelişimi olmuş ve% 83.33'ünün virüs(ler)ten ari olduğu tespit edilmiştir. Latona ve Marfona çeşidine ait fidelerde PVY, Sante çeşidinde ise PLRV bulunduğu saptanırken, virüs arındırma oranları sırası ile %69.56, %68.42 ve %70.83 düzeyinde olmuştur. Nod kültürü ile çoğaltılan ve virüsten ari olan 444 adet fide toprağa aktarıldığında, Marabel ve Milva çeşitleri dışındaki 9 çeşitten toplam 223 adet fide (%50.22) gelişme göstermiş ve bunların 143 adedi (%64.12) mini yumru oluşturmuştur. Çeşitlere göre değişen miktarlarda toplam 462 adet mini yumru elde edilmiş ve yapılan DAS-ELISA testi sonucunda bu yumruların PVX, PVY, PLRV, PVS ve PVM adlı virüsleri içermediği saptanmıştır.
