Kargı ve Osmancık Çevresindeki Yayla Topraklarının Aktinomiset Çeşitliliği

Abstract
Bu çalışmada Çorum iline bağlı Kargı (Eğinönü), Abdullah ve Osmancık-Başpınar Karaca yaylalarından alınan toprak örneklerinden izole edilen aktinomiset suşlarının 16S rRNA gen bölgesi analizlerine göre aktinomiset çeşitliliğinin saptanması amaçlandı. Dilüsyon plak tekniğinin uygulandığı izolasyon çalışmasında 5 farklı seçici besiyeri kullanıldı. İzolatların beş farklı kültür ortamında kültürel karakterizasyonları yapıldı. Test izolatlarının 16S rRNA gen bölgesinin PZR amplifikasyonu 27F ve 1525R primerleri ile gerçekleştirildi. Filogenetik ağaçlar, MEGA 7.0 yazılım programında neighbor-joining algoritmasıyla oluşturuldu. 5 farklı izolasyon besiyerinden toplam 15 mikroorganizma seçildi ve 16S rRNA gen dizi analizine göre 15 izolatın, altısı Nocardia cinsi üyesi, beşi Streptomyces cinsi üyesi, biri Haloactinopolyspora cinsi üyesi, biri Micromonospora cinsi üyesi, biri Nocardiopsis cinsi üyesi ve birinin de Rhodococcus cinsi üyesi olduğu tespit edildi.

Abstract
In this study, it was aimed to determine the diversity of actinomycetes according to 16S rRNA analysis of actinomycete isolates obtained from soil samples taken from Kargı (Eğinönü), Abdullah and Osmancık-Başpınar Karaca plateaus in Çorum Province. The isolates obtained were isolated using a dilution plate technique on 5 different selective media. Cultural characterizations of the isolates were carried out in five different culture media. PCR amplification belonging to 16S rRNA gene region of the isolates was realized out with primers of 27F and 1525R. Phylogenetic trees were formed with the neighbor-joining algorithm in MEGA 7.0 software. A total of 15 microorganisms were selected from 5 different isolation media and according to 16S rRNA gene sequence analysis of 15 isolates, six Nocardia isolates, five Streptomyces isolates, one Haloactinopolyspora isolate, one Micromonospora isolate, one Nocardiopsis isolate and one Rhodococcus isolate were determined.
