Çanakkale’de Limoni Servilerde Kanser Etmeni Seiridium cardinale’nin Yaygınlığının Saptanması, Morfolojik ve Patojenik Karakterizasyonu

Abstract

Servi kanserine sebep olan Seiridium cardinale (W.W. Wagener)’nin Çanakkale’de limoni servilerde (Cupressus macrocarpa cv. Goldcrest) yaygınlığı, hastalık oranı, morfolojik ve patojenik karakterizasyonun saptanması amacıyla 2021-2022 yılları arasında sörvey gerçekleştirilmiştir. Etmenin sörvey alanının tamamında var olduğu ve hastalık oranının % 1.65 olduğu saptanmıştır. Ana gövde veya yan dallarda geriye doğru ölüm, dal veya gövdede kanser oluşumu, kabuk dokusunda çatlama ve zamklanma ile acervulus oluşumu hastalığın teşhisinde önemli belirtiler olmuştur. Etmenin 5 farklı fungal besin ortamında ve 3 farklı sıcaklık derecesinde inkübe edilmesiyle, koloninin en iyi 250C’de malt ekstrakt agar (MEA) ve patates dekstroz agarda (PDA) geliştiği saptanmıştır. Fungusun PDA besin ortamında kirli beyaz, gri renk koloni oluşturduğu, koloninin altının ise pembemsi turuncu, somon renkli olduğu gözlemlenmiştir. Patojenisite çalışmalarında test edilen izolatların tamamının virülent olduğu saptanmıştır. Çanakkale’de tercih edilen diğer koniferlere olan etkisini araştırmak amacıyla yapılan çalışmada Cupressocyparis leylandii, Cupressus arizonica cv. Greene ve Cupressus sempervirens cv. Strict’de hastalık simptomları gözlenmiş, fakat Thuja occidentalis cv. Smaragd, Thuja orientalis cv. Aurea compacta Nana ve Pinus pinea fidanlarında simptom oluşumu gözlenmemiştir. Bu sonuçlar Türkiye’de servi kanserinin limoni servilerde yaygınlığını, morfolojik ve patojenik karakterizasyonunu rapor eden ilk çalışma niteliğindedir.
