Investigatıon of Citrus exocortis viroid (CEVd) in Ornamental Citrus Plants
Abstract
Citrus fruit production is one of the most important agricultural branches in our country. A large portion (approximately 70%) of the 3.6 million tonnes is produced in the Eastern Mediterranean region. There are many factors that negatively effect the production of citrus fruit. One of these factors is viroid and virus diseases. In recent years, the production of ornamental citrus in our region has been rapidly increasing and creating a new market. In this study, the existence of Citrus exocortis viroid (CEVd),one of important viroid agent in Eastern Mediterranean citrus production, was investigated. The study was carried out in citrus varieties of Kumquat (Fortunella japonica), Nippon mandarinquat (C. sinensis x C. Japonica) and Calomandin (Citrofortunella microcarpa). Three of each variety were selected and the presence of disease agents in these varieties was investigated by biological indexing and molecular methods. Molecular studies were carried out using the Reverse Polymerase Chain Reaction (RT-PCR) method in which Gynura aurantica and Etrog citron (C. medica L.) plants were used as indicator plants in biological indexing studies. It was determined that all the ornamental citrus varieties examined were contaminated with CEVd. As a result of indexing studies, symptom developments such as severe leaf epinasty, shortening between nodes, stunting, development of necrosis on the back side of the leaves were observed in Etrog citron plants and leaf epinasty, shortening between the nodes ,stunning symptoms and necrosis were observed in Gynura aurantica. As a result of RT-PCR studies, 371 nt long bands of CEVd were determined by agar gel electrophoresis.
