Investigation of Babesia bovis and Babesia bigemina by Real Time PCR in Tick Species Collected from Cattle in Kayseri Province

Abstract

This study was performed to investigate Babesia bigemina and B. bovis via real time PCR analyses in ge-nomic DNA pools constructed from adult ixodid ticks on cattle in different parts of Kayseri, and to detect the phyloge-netic characters of the isolates. TaqMan and Sybergreen real time PCR analyses with rap-1a and msa-2c gene specific primers for B. bigemina and B. bovis were performed in DNA pools of 1115 adult ticks. B. bovis and B. bigemina were detected in two (5.2%) and eight (21%) out of 38 DNA pools. No mix infection was determined. B. bigemina was found in four Rhipicephalus annulatus, two R. turanicus, one Hyalomma marginatum, and one H. anatolicum pools whereas both two positive B. bovis samples were detected in H. marginatum pools. Ct (cycle threshold) (dR) values were deter-mined as 35.75 and 37.71 for B. bigemina and B. bovis. Two B. bigemina positive samples were sequenced and de-posited into GenBank (KU680419-KU680420). In phylogenetic analyses of BbigrapKys1 and BbigrapKys2 isolates, 100% identity amongst themselves and 1±0.5% and 0.7±0.4% genetic distances were found with other similar isolates from the world and Turkey. In conclusion, the scientific knowledge was obtained about the molecular prevalence of B. bigemina and B. bovis in ticks collected from cattle in Kayseri province and also about molecular characterization and phylogenies of some positive isolates.

Kayseri Yöresindeki Sığırlardan Toplanmış Kene Türlerinde Babesia bovis ve Babesia bigemina’nın Real Time PCR Yöntemiyle Araştırılması*

Abstract

Bu çalışma, Kayseri’nin farklı lokalizasyonlarındaki sığırlardan toplanmış erişkin ixodid kenelerden oluşturulmuş genomik DNA havuzlarında Babesia bigemina ve B. bovis’in real time PCR analizleriyle araştırılması ve izolatların filo-genetik karakterlerinin saptanması amacıyla gerçekleştirilmiştir. Sığırlardan toplanmış 1115 erişkin ixodid keneden oluşturulmuş DNA havuzlarında, B. bigemina ve B. bovis için rap-1a ve msa-2c gen bölgelerini amplifiye eden primer-lerle TaqMan prob bazlı ve Sybergreen tabanlı real time PCR analizleri yapılmıştır. Toplam 38 genomik DNA havuzu-nun ikisinde (%5.2) B. bovis ve sekizinde (%21) B. bigemina pozitifliği saptanmıştır. Miks enfeksiyon belirlenmemiştir. Babesia bigemina pozitiflerin dördü Rhipicephalus annulatus, ikisi R. turanicus, biri Hyalomma marginatum ve biri H. anatolicum havuzlarında, B. bovis pozitiflerin ikisi de H. marginatum havuzunda saptanmıştır. Ct (cycle threshold) (dR) değerleri B. bigemina ve B. bovis için ortalama 35.75 ve 37.71 belirlenmiştir. Moleküler karakterizasyon amacıyla nes-ted PCR analizleriyle uygun DNA bant profilleri gösteren B. bigemina pozitif sekiz örnekten ikisi sekanslanmış ve Gen-Bank kayıtları (KU680419-KU680420) gerçekleştirilmiştir. BbigrapKys1 ve BbigrapKys2 izolatlarının filogenetik analizle-rinde kendi aralarında %100 identiklik, Dünyadaki ve Türkiye’deki diğer benzer izolatlarla %1±0.5 ve %0.7±0.4 genetik farklılık tespit edilmiştir. Sonuç olarak bu çalışmayla, Kayseri yöresindeki sığırlardan temin edilmiş kenelerde B. bigemi-na ve B. bovis’in varlığı ve moleküler prevalansı ile pozitif bazı izolatların moleküler karakterizasyonu ve filogenileri hakkında bilimsel veriler elde edilmiştir.
