KAYNAK ALABALIĞI (SALVELINUS FONTINALIS)’NIN SPERMATOLOJİK ÖZELLİKLERİ VE SPERMANIN KISA SÜRELİ MUHAFAZASI

Abstract

Bu çalışma kaynak alabalığının spermatolojik özelliklerini belirlemek ve spermanın kısa süreli muhafazası için sulandırıcı geliştirmek amacıyla yapılmıştır. Üreme sezonunda 6 erkek bireyden sperma abdominal masaj yöntemi ile alınmış ve spermalarda, miktar, motilite, motilite süresi, spermatokrit oranı, yoğunluk ve pH belirlenmiştir. Kaynak alabalığı spermalarında; miktar (ml), motilite (%), motilite süresi (s), spermatokrit oranı (%), yoğunluk (x 109 spermatozoon/ml) ve pH değerleri sırasıyla ortalama 4.3 ± 1.54, 98.3± 4.08, 59.2 ± 17.27, 27.1 ± 4.47, 12.9 ± 1.29 ve 6.9 ± 0.15 bulunmuştur. Sulandırıcı S1’de muhafaza edilen spermanın daha yüksek bir motiliteye sahip olduğu ve sulandırıcı S1’in diğerinden daha etkin olduğu görülmüştür.

Abstract

This study was carried out to determine the spermatologic characteristics and to develop an extender for the short-term storage of semen from brook trout. Four different extenders were tested in the study: a) 8.75 g/l NaCl, 0.20 g/l KCl, 0.20 CaCl2, 0.30 g/l NaHCO3 (S1), b) 8.75 g/l NaCl, 0.20 g/l KCl, 0.20 MgCl2, 0.40 g/l NaHCO3 (S2), c) 7.25 g/l NaCl, 0.40 g/l KCl, 0.80 g/l NaHCO3, 2.0 g/l glucose (S3), d) 8.75 g/l NaCl, 0.20 g/l KCl, 0.10 CaCl2, 0.10 g/l MgCl2, 0.40 g/l NaHCO3 (S4). In the spawning season, sperm was collected by abdominal massage from 6 males. In collected sperm; volume, motility, duration of motility, spermatocrit, density and pH were determined. In the sperm of brook trout, volume (ml), motility (%), duration of motility (sec), spermatocrit, density (x 109spermatozoon/ml), and pH values were found as mean 4.3 ± 1.54, 98.3± 4.08, 59.2 ± 17.27, 27.1 ± 4.47, 12.9 ± 1.29 ve 6.9 ± 0.15, respectively. Sperm stored in extender S1 had a higher rate of motility and the extender S1 seemed to be more effective than the others
