Complexation of Docetaxel in Cyclodextrin Derivates With Different Methods and Characterization of Inclusion Complexes

Abstract

In present study, cyclodextrin-docetaxel complexes containing active ingredient called docetaxel that shows anticancer effect were prepared. In order to compare the obtained complexes and preparation methods, two different cyclodextrin derivates (hydroxypropyl-betacyclodextrin (HP-β-CD) and methyl-beta-cyclodextrin ( M - β - C D ) ) w i t h f i x e d m o l a r r a t i o (cyclodextrin:docetaxel 1:2) and two different methods were used and totally four different complexes were prepared. Phase solubility studies, morphological structure determination with scanning electron microscope (SEM), differential scanning calorimeter (DSC) and fourier transform infrared spectroscopy (FTIR) analyses of these complexes were made. When all these studies were evaluated together, it was a concluded that docetaxel could form complexes with both cyclodextrin derivates and optimum results are obtained with freeze-drying method and methyl-βcyclodextrin.

DOSETAKSELİN SİKLODEKSTRİN TÜREVLERİ İLE FARKLI YÖNTEMLERLE KOMPLEKS OLUŞTURMASI VE OLUŞAN İNKLÜZYON KOMPLEKSLERİNİN KARAKTERİZASYONU

Abstract

Bu çalışmada, antikanser etki gösteren dosetaksel etkin maddesini içeren siklodekstrin-dosetaksel inklüzyon kompleksleri hazırlanmıştır. Elde edilen kompleksleri ve hazırlama yöntemlerini karşılaştırabilmek amacıyla, sabit molar oranda (siklodekstrin:dosetaksel/1:2) iki farklı siklodekstrin türevi [hidroksipropopil-betasiklodekstrin (HP-β-CD) ve metil-beta-siklodekstrin (Mβ-CD)] ve iki farklı yöntem (birlikte dondurarak kurutma ve yoğurma) kullanılmış, toplamda dört farklı kompleks hazırlanmıştır. Elde edilen bu inklüzyon komplekslerinin faz çözünürlüğü çalışmaları, taramalı elektron mikroskobu (SEM) ile morfolojik yapı tayinleri, diferansiyel taramalı kalorimetre (DSC) ve fourier dönüşümlü infrared spektrofotometre (FT-IR) analizleri gerçekleştirilmiştir. Tüm bu çalışmalar birlikte değerlendirildiğinde, dosetakselin her iki siklodekstrin türevleri ile başarıyla kompleks oluşturabildiği ve optimum sonuçların ise metil-β-siklodekstrin ve birlikte dondurarak kurutma yöntemi kullanılarak elde edildiği sonucuna varılmıştır.
