İNSAN KAN LENFOSİTLERİNDE SİTRİNİNİN MİTOTİK İNDEKS ÜZERİNE ETKİSİNİN ARAŞTIRILMASI

Abstract

Mikotoksinler, insanlarda ve hayvanlarda hastalıklara ve ölüme sebep olabilen mikrofunguslar tarafından oluşturulan ikincil metabolitlerdir. Bu çalışmada, insan periferal kan lenfositlerinde mikotoksin sitrininin (CTN) mitotik indeks (MI) üzerine etkileri araştırılmıştır. Altı sağlıklı sigara içmeyen bireyden (3 erkek ve 3 kadın) heparinize periferal kan örnekleri alınmış ve Nutrient Mixture F10 kültür ortamında 72 saat süre ile her bir kişi için 9 ayrı tüpte kültüre edilmiştir. Kültür işleminin son 48 saati boyunca, lenfosit kültürleri 10 mM, 20 mM, 40 mM, 60 mM, 80 mM, 100 mM konsantrasyonlarda CTN, 0.1 mM konsantrasyonda mitomisin c (pozitif kontrol) ve % 0.5 absolu etanol ile muamele edilmiştir. Metafaz elde etmek için, kültürlere 70. saatte 0.1 μg/ml son konsantrasyonda kolsişin eklenmiş ve 72. saatte kültür işlemi sonlandırılmıştır. 60 mM, 80 mM ve 100 mM CTN konsantrasyonlarındaki MI değerlerinin, negatif kontrollerin MI değerlerine göre önemli bir şekilde azaldığı bulunmuştur (p=0.001). Ayrıca, CTN konsantrasyona bağlı olarak da insan lenfositlerindeki MI oranlarında önemli bir azalmaya sebep olmuştur. Bu sonuçlar, kültüre edilmiş insan lenfositlerinde yüksek konsantrasyonlardaki CTN’in sitotoksik olduğunu düşündürmektedir

Investigation of Effect of the Citrinin on Mitotic Index in the Human Blood Lymphocytes

Abstract

Mycotoxins are secondary metabolites produced by microfungi that are capable of causing disease and death in humans and other animals. In the present study, the mycotoxin citrinin (CTN) was evaluated for their effects on mitotic index (MI) rates of human peripheral blood lymphocytes. Heparinized peripheral blood samples were obtained from six healthy non-smoking individuals (3 females and 3 males) and cultured for 72 h in the culture medium Nutrient Mixture F-10 in 9 different tubes for each individual. Lymphocyte cultures were treated for the last 48 h with CTN at the following concentrations, 10 mM, 20 mM, 40 mM, 60 mM, 80 mM, 100 mM, and 0.1 mM mitomycin c (positive control) and 0.5% absolute ethanol. To block metaphases, at 70 h of incubation, colchicine was added to cultures to give a final concentration of 0.1 μg/ml, and cultures were harvested cells at 72 h. At 60 mM, 80 mM and 100 mM CTN concentrations, significantly reduced MI values in comparison with negative controls were found (p=0.001). In addition, CTN caused a significant concentration-dependent decrease in MI rates in human lymphocytes. These results make thinking that CTN at the high concentrations is cytotoxic in cultured human lymphocytes
