The Investigation of Cobalt Chloride effects on Some Toxicokinetic Parameters of Cyanide and Thiocyanate

Abstract

This study was made to determine the same toxicokinetic parameters of eyanide and thiocyanate in sheep experimentally poisoned with amygdalin. In this study. LS sheep 50 - 60 kg weights were used. Amygdalin and cobalt chloride were given and the eyanlde and thioeyanate levels in samples were determined Then some ıoxicokinetic pa­ramelers of Ihe eyanide and Ihioeyanaıe in the blood samples laken al 0.5. 1, 2. 4. 8, 24. 48 and 72 ın hours were es­tablished. The cobalt chloride was given intravenously in doses 5 and İS mçVkg and then the distribution half-lives of blood eyanide levels according to doses were respectively, 3.17±O.35 and 4.03±O.65 hours; the elimination half-lives were respectively, 27.62±3.16 and 39.53±5.35 hours; mean residence times were respectively, 44.44±2.76 and 109.29±8.43 hours; Ihe duratlon of maximat coneentration were respectively, 241h hours; the maximal blood levels were respectively, 0.120 and 0128 �glml were determined. Besides, area under the curve values according to doses were respectively, 7.09±1.24 and 5.93±O.64 �g. hour/ml; volumes of distribution at steady state were respectively, 0.099±O.OO5 and 0.125iO.Ol l/kg; clearanee values were respeclively, 1.21iO.44 and 2.52±1.21 mVhour. kg were cal­culated. The distribution half-Ilves of blood thioeyanaıe levels according lo doses were respeclively. 0.30±0.02 and 2.49±1.12 hours; the eliminatian half-lives were respectively, 213.89±11.21 and 96.56±9.35 hours; mean residence times were respectively, 309.09±25.76 and 142.95±15.66 hours; the duralian of maximal concentration were res­pectively, 2 and ılh hours; the maximal blood levels were respectively, 0.028 and 0.032 �glml were determined. Be­sides, area under the curve values according lo doses were respeclively, 8.77±1.44 and 3.26t1.5S J.lg. hour/ml; vo­lumes of distribution al steady state were respectively, 0.211iO.Ol and 1.333iO.05 l/kg;; cleararıce values were respectively. 0.56iO.23 and 7.66±1.55 mVhour. kg were established

SİVANÜR ve TİYOSİYANATIN BAZI TOKSİKOKİNETİK PARAMETRELERİ ÜZERİNE KOBALT KLORÜRÜN ETKİSİNİN ARAŞTIRILMASI

Abstract

Bu çalışma deneysel olarak amigdalinle zehinenen koyunlarda kan siyanür ve tiyosiyanat düzeylerine ail bazı toksikokinetik parametreler üzerine kobalt kIorOrOn etkilerini araştırmak amacıyla yapıldı. Bu çalışmada. ağırlıkları 5060 kg arasında olan IS koyun kullanıldı. Amigdalin ve kobalt klorür verildikten sonra 0.5,1,2,4,8,24,48 ve 72. sa­atlerde alınan kan örneklerindeki siyanür ve liyosiyanat düzeylerine ait bazı toksikokinelik parametreler belirlendi. Ko­balt klorür, 5 ve 15 mglkg dozlarında damar içi yolla verildi. Daha sonra kan siyanür düzeylerine ait da Jılım dönemi yarı ömünerinin dozlara göre sırasıyla, 3.17±O.35 ve 4.03±O.65 saat; atılım dönemi yarı ömürlerinin sırasıyla, 27.62±3.16 ve 39.53±5.3S saaı; ortalama kalış sürelerinin sırasıyla, 44.44±2.76 ve 109.29±8.43 saat; doruk yo{ıunluğa ulaşma sürelerinin 24. saat; doruk siyanür düzeylerinin ise sırasıyla, 0.120 ve 0.128 J.lglml olduğu tespit edildi. Bunun yanında, siyanür düzeylerine ait e Jri altında kalan alan değerleri sırasıyla, 7.Q9±1.24 ve 5.93±O.64 J.lg saat/ml; karanı dun.ımdaki dağılım hacimleri sırasıyla, 0.099±0.OO5 ve 0.125±O.Ol Ukg; klirens de{jerleri ise sırasıyla. 1.21:tD.44 ve 2.52±1.21mVsaaı. kg olarak hesaplandı. Kan tiyosiyana düzeylerine ait dağılım dönemi yarı ömürlerinin dozlara göre sırasıyla, 0.30±0.02 ve 2.49±1.12 saat; atılım dönemi yarı ömürlerinin sırasıyla, 213.89±11.21 ve 96.56±9.35 saat; or­talama kalış sürelerinin sırasıyla. 309.09±25.76 ve 142.95±15.66 saat; doruk yo{ıunluğa ulaşma sürelerinin sırasıyla, 2 ve I. saal; doruk tiyosiyanaı düzeylerinin ise sırasıyla,' düzeylerinin eğri altında kalan alan değerleri sırasıyla. 8.77±1.44 ve 3.26±1.55 �g. saaVml; karanı durumdaki dağılım hacimleri sırasıyla, 0.21IiO.01 ve 1.333iO.05 Ukg; klirens değerlerinin ise sırasıyla 0.56±O.23 ve 7.66t1.55 mVsaat. kg olarak tespit edildi
