Abstract

After food and mouth disease vaccirte preparation safety and protaction
controls have to be done to be su re about its quality and reliability.
This tests can be done either in vivo such as antibody free susceptible
calves, suckling mice, guinea pigs or in vitro such as tissue cultures. For
that purpose the most practicle economical and reliable systems must be
prefered. Guiriea pigs and tissue culture systems are böth more practicle
and economical than cattle tests.
In this work those last two test systems have been studied comperatively
to find out the most sensitive and practical one. ı:or that purpose 1 O
series of BEl inactivated alhydrogel A22 and o, type FMD vaccines have
been tested for their inıiocuity by using 21 O guinea pigs and BHK-21 monolayer
tissue culture. The presence of trace infective virus particules have
been detected by the vesicles on the guinea pig pads and CPE in tissue
cultures and have been confirmed by complement fixation test (CFT).
At the end it was concluded that although the guinea pig tests are
mo re practicle and economical. The tissue. culture tests are the most reliable
for safety controls of inactivated FMD vaccines.

Şap Aşılarının Kobaylarda ve Doku Kültüründe Zararsızlık Kontrolü

Abstract

Şap aşılarının hazırlanmasından sonra, zararsızlık ve bağışıklık kontrolleri
yapılarak, aşının kalite ve güvenilirliğinden emin olunur. Bu testler
antikorsuz duyarlı danalar, yavru fareler, kobaylar gibi invivo veya doku
kültürü gibi invitro sistemlerde yapılabilir. Pratik, ekonomik ve güvenilir sistem
tercih edilmelidir. Bu çalışmada kobay ve doku kültürü testleri duyarlılık
ve uygulamadaki kolaylıklar yönünden karşılaştırmalı olarak çalışılmıştır.
Bu amaçla 21 O kobay ve BHK-21 manolayer hücre kOltOrO kullanılarak BEl
ile inaktive aluminyum jeline adsorbe 1 O sed A22 ve o, tipi şap aşılarınin
zararsızlık kontrolü karşılaştırmalı olarak çalışılmıştır. Enfektif virus partiküllerinin
varlığı kobay tabanında vesikül oluşumu, doku kültüründe CPE
ile belirlenmiş , Complement Fiksasyon Test (CFT) ile doğrulanmıştır.
Sonuçta kobay testleri pratik ve ekonomik olmasına rağmen, doku
kültürü. testleri inaktiv şap aşılarının zararsızlık kontrolünde daha güvenliir
bir testtir.
