Effectiveness of Native Trichoderma Isolates Against Gray Mold in Vitis vinifera

Abstract
Gray mold is a significant fungal disease in grapevine cultivation, causing by Botrytis cinerea. This study aimed to evaluate the antagonistic effect of eight native Trichoderma isolates against B. cinerea. In vitro assessments using dual culture tests demonstrated mycelial growth varying degrees of inhibition. Trichoderma isolates inhibited the mycelial growth of B. cinerea by 68.0-79.4%. Five Trichoderma isolates (Tr91, Tr52, Tr142, Tr117, and Tr93) showed high effect against B. cinerea. In vivo experiments were executed to further assess antifungal efficacy. Trichoderma spore suspension were collected after ten days incubation in potato dextrose agar (PDA). Spore concentration of Trichoderma was adjusted to 1×109 spore/mL. B. cinerea was growthed five days in PDA for acquiring conidial suspension. Conidia suspension of B. cinerea (1×10⁶ conidia/mL) was first sprayed to the leaves and bunches with a custom-built spray tower at 1 atm pressure, followed two hours later by Trichoderma spore suspension (spraying 2 mL per plant tissues). Control groups included B. cinerea only (positive), and a sterile distilled water only (negative). Results in the leaves test showed that Trichoderma sp. Tr52 significantly reduced disease severity of B. cinerea by 51.3% compared to the positive control. Trichoderma sp. Tr52 had an exhibit highest efficacy (50.7%) against gray mold disease in bunches. These findings suggest that native Trichoderma isolates have potential as a biological control against gray mold on grapevine. Future research should focus on evaluating their effectiveness against different fungal pathogens of limiting grapevine cultivation.
