Investigation of Amounts of Malondialdehyde, Glutathione and Vitamins with Total Antioxidan Capacity in Plant Mentha pulegium L.

Abstract

In this study, the amounts of malondialdehyde (MDA), reduced form glutathione 1), riboflavin (vitamin B2), nicotinic acid (vitamin B3), pyridoxine hydrochloride (vitamin B6) and folic acid (vitamin B9) in leaves of Mentha pulegium L. sample were determined by using High Performance Liquid Chromatography. Also, total phenolic and flavonoid contents of plant samples were determined by spectrophotometric measurements. In addition to these, the free radical scavenging effect of the plant extract on 2,2-diphenyl-1-pikrilhidrazil (DPPH), this effect was measured using the standard the antioxidant known butylated hydroxytoluene (BHT) and compared. The amount of MDA, GSH, GSSG and vitamin A, vitamin E, β-carotene, vitamin C, vitamin B1, vitamin B2, vitamin B3, vitamin B6 and vitamin B9 in leaves of Mentha pulegium L. were obtained to be 17,43±1,57g/g; 185,71±10,61 g/g; 280,48±24,58 g/g; 0,38±0,08 g/g; 12,46±1,82 g/g; 1,24±0,14 g/g; 282,86±9,24 g/g; 0,50±0,09 g/g; 43,11±7,02 g/g; 16,91±1,59 g/g; 81,88±9,47 g/g and 1,53±0,16 g/grespectively. The amount of total phenolic the as 136,85±11,39 (mg/g gallic acid equivalent) and the amount of the total flavanoid the 65,2±1,07 (mg/g quercetin equivalent) in samples were determined. Free radical scavenging effect of 85,94±1,20 % was determined as, but compared to BHT was not significant difference (p> 0.05). It may be concluded from the results that rich, the the Mentha pulegium L. plant contains of glutathione (GSH, GSSG), vitamin C, vitamin B2 and vitamin B6, also amount contains phenolic and flavonoid content a powerful antioxidant

Yarpuz (Mentha pulegium L.) Bitkisindeki Malondialdehit, Glutatyon ve Vitamin Miktarları ile Total Antioksidan Kapasitesinin Araştırılması

Abstract

Bu çalışmada, yarpuz (Mentha pulegium L.) bitkisinin yapraklarındaki malondialdehit (MDA), indirgenmiş glutatyon (GSH), yükseltgenmiş glutatyon (GSSG) ile A vitamini, E vitamini, β-karoten, C vitamini, tiamin klorür (B1 vitamini), riboflavin (B2 vitamini), nikotinik asit (B3 vitamini), pridoksin klorür (B6 vitamini) ve folik asit (B9 vitamini) vitaminlerinin miktarları Yüksek Performanslı Sıvı Kromatografisi (HPLC) ile belirlendi. Ayrıca bitki örneğinin toplam fenolik ve flavonoit miktarları spektrofotometrik ölçümlerle tespit edildi. Bunlara ek olarak bitki ekstraktının serbest radikal temizleme etkisi 2,2-difenil-1-pikrilhidrazil (DPPH) kullanılarak ölçüldü ve bu etki standart antioksidanlığı bilinen bütillenmiş hidroksitoluen (BHT) ile kıyaslandı. Yarpuz bitkisinin yapraklarındaki MDA, GSH, GSSG, A, E, β-karoten, C vitamini, B1, B2, B3, B6 ve B9 vitaminlerinin miktarları sırası ile 17,43±1,57ï­g/g; 185,71±10,61 ï­g/g; 280,48±24,58 ï­g/g; 0,38±0,08 ï­g/g; 12,46±1,82 ï­g/g; 1,24±0,14 ï­g/g; 282,86±9,24 ï­g/g; 0,50±0,09 ï­g/g; 43,11±7,02 ï­g/g; 16,91±1,59 ï­g/g; 81,88±9,47 ï­g/g ve 1,53±0,16 ï­g/g olduğu gözlendi. Örneklerdeki toplam fenolik miktarı 136,85±11,39 (mg/g gallik asit eşdeğeri), toplam flavonoit miktarı ise 65,2±1,07 (mg/g kuersetin eşdeğeri) olarak belirlendi. Serbest radikal temizleme etkisi ise % 85,94±1,20 olarak belirlendi, fakat BHT'ye kıyasla anlamlı fark bulunamadı (p>0.05). Sonuç olarak elde edilen bulgulardan Mentha pulegium L. bitkisinin, Glutatyon (GSH, GSSG), C vitamini, B2 ve B6 vitaminleri açısından iyi bir kaynak olduğu, içerdiği zengin fenolik ve flavonoit içeriği göz önüne alındığında ise güçlü bir antioksidan bitki olduğu söylenebilir.
