Özet

Bir bitki büyüme hormonu olan oksin Indol-3-Asetik Asit'in (IAA) reaktif oksijen türlerini (ROT) ürettiği, nötrofil, lenfosit ve makrofajlar üzerine sitotoksik etkisi olduğu bilinmektedir. Bu çalışmada, IAA'in üçüncü nesil farelerin (Mus musculus) kemik iliği hücreleri mitotik indeksine etkisinin araştırılması amaçlanmıştır. Bu amaçla, dişi fareler IAA, kontrol etanol ve kontrol serum fizyolojik grubu olmak üzere üç gruba bölündü. İki dişi bir erkekle çiftleştirildi. IAA, farelere 300 mg/kg vücut ağırlığı dozu 1/40 sulandırılarak üç günde bir deri altına (ip) verildi. Kontrol etanol grubuna IAA'nın çözündüğü etanol ve kontrol serum fizyolojik grubuna da serum fizyolojik aynı yolla verildi. Bu işlemler üç nesil boyunca tekrarlandı. Çalışma sonunda, IAA'in üçüncü nesil farelerde mitotik indeksi, kontrol grubuna göre arttırdığı gözlendi.

Sonuç olarak, IAA'in mitotik indeksi aktive ettiği söylenebilir.

Abstract



The effect of indole-3-acetic acid on mitotic index on bone marrow cells of third generation mice

The plant growth-hormone auxin indole-3-acetic acid (IAA) is known to generate reactive oxygen species and be cytotoxic on neutrophils, macrophages and lymphocytes. The aim of present study was to investigate the effect of IAA on mitotic index in the bone marrow cells of third offsprings mice (Mus musculus). Female mice were divided into three groups: IAA-treated group and ethanol control group and serum physiologic control group. For mating, two females and one male were held in a steinless steel cage. All chemicals were applied intraperitoneally (i.p.). IAA was administered to maternal mice as a 1/40 diluation of 300 mg/kg body weight in 3 day intervals. Ethanol and serum physiologic was administered in the control group. These applications were continued for three generations.

Mitotic index increased in the IAA groups compared to control groups in the bone marrow cells of the third generations mice.

The results demonstrated that IAA increased mitotic index in the bone marrow cells of the third generations of mice.
