In vitro Adventitious Shoot Regeneration from Leaves and Petiols of Gerbera (Gerbera jamesonii Bolus)
Abstract
In this study, effects of plant growth regulators on shoot regeneration from leaf and petiol
explants of Gerbera cultivar ‘Rosalin’ were determined. For this purpose, 18 different combinations of
plant growth regulators consisting of three doses of AgNO3 (0, 1.0 and 2.0 mg/L) and three doses of
BAP (1.0, 2.0 and 3.0 mg/L) or thidiazuron (0.3, 0.6 and 1.2 mg/L) were tested. In addition, 0.1 mg/l
NAA was added to all MS media. In the study, TDZ applications gave better results based on the
formation of callus for both explants types. When the leaves were used as explants, no shoot
formation was observed on MS medium containing both BAP and TDZ. When the petiole was used as
explants, BAP applications gave better results than TDZ application in term of shoot regeneration. In
the study, the highest shoot regeneration rate, shoot number and leaf number were 22.5%, 2.0 shoots,
and 3.33 leaves on the MS medium containing 3 mg/l BAP + 0.1 mg/l NAA, respectively.
Applications of silver nitrate were not effective on in vitro shoot regeneration in gerbera.
