Ultrastructural Demonstration Of The Effects Of Streptozotocin-Induced Diyabetes On Heart Muscle In Female Rats

Abstract

The aim of this study was to analyze the ultrastructural alterations in ventricular cardiac muscle cells in female rats in which chronic diabetes was induced with streptozotocin. Mature female rats (n=18) were assigned into 3 groups: Group 1: the controls (n=6), Group 2: the sham controls (n=6), and Group 3: the diabetic (n=6). Eight weeks after injection, left ventricle muscles were extracted and examined with a LEO 906E transmission electron microscope. The electron microscopic analyses of the left ventricle wall revieled prominent degenerative alterations in the myocardial cells of diabetic rats. Swelling and crystolysis in the mitochondria, lipid deposition in the cytoplasm, and widespread effecement of myofilaments were detected. The Z bands were irregular and effacement was noted. In edition, the intercalated discs were dilated. As a result, it can be suggest that females are affected at the early stages of diabetes mellitus.

Dişi Sıçanlarda Streptozotosin İle Oluşturulmuş Diyabetin Kalp Kası Üzerine Etkisinin Ultrastrüktürel Düzeyde Gösterilmesi

Abstract

Çalışmada streptozotosin ile kronik diyabet oluşturulmuş dişi sıçanlarda ventriküler kalp kasının ultrastrüktürel düzeyde incelenmesi amaçlanmıştır. Çalışmada 18 adet dişi erişkin sıçanlardan 3 grup oluşturulmuştur. 1. grup: kontrol (n=6), 2. grup: sham kontrol (n=6), 3. grup: diyabet (45mg/kg streptozotosin) (n=6). 8 hafta sonra diyabet ve kontrol gruplarından sol ventrikül bölgeleri alınarak, LEO 906E transmission elektron mikroskopta incelenmiştir. Diyabetik grubun kas hücrelerinde belirgin dejeneratif değişiklikler saptanmıştır. Mitokondrionlarda belirgin şişkinlik ve kristolizis, sitoplazmada yaygın lipid birikimi, miyoflamanlarda yaygın silinme dikkati çekmiştir. Z bantlarında düzensiz seyir ile silinme izlenmiştir. Ayrıca interkalat disklerde dilatasyon ilgiyi çekmiştir. Bulgularımız ışığında yapılan değerlendirmede, kadınların diyabetten erken dönemde etkilendiği sonucuna varılmıştır.
