Effects of L-Arginine and CaEDTA in Temporal Lobe Epilepsy

Abstract

This study aimed to assess the effect of L-Arginine, a nitric oxide (NO-) procursor, and CaEDTA, an extracellular zinc chelator, on the short time epileptic seizure which was formed by pilocarpine HCl injection. The experiment was consist of 4 groups, including seizure control (saline 10 µl, i.c.v., and then 380 mg/kg pilocarpine HCl i.p.), L-Arginine (150 µg/10 µl, i.c.v.), CaEDTA (100 mM, 10 µl, i.c.v.), L-Arginine+CaEDTA groups. Seizure behaviour of the rats were observed for two hours following the injections and the animals were sacrificed at the end of the process and the brains were taken out for immersion Autometallography (iZnSAMG, NeoTimm) and N-6- methoxy-8-quinolyl-para-toluenesulfonamide (TSQ) staining processes. Seizure behaviour in the groups of seizure control, L-Arginine, CaEDTA and L-Arginine+CaEDTA was found to be strong and equal to each other. Moreover, in these groups no zinc translocation was spotted in hippocampal stainings which were performed by using zinc histochemistry. With these results, we concluded that the NOprocursor injection alone, zinc chelation alone or together had no effects especially on seizure activity and zinc depletion in the hippocampus during and at the end of the two hour epileptic seizure.
