Comparison of HPLC, CMIA and ECLIA Methods for Measurement of Plasma 25-OH Vitamin D
Abstract
The aim of the study is to compare two automated chemiluminescent immunassays [Chemiluminesence immunoassay (CMIA) and electro-chemiluminescent immunoassay (ECLIA)] with the high performance liquid chromatography (HPLC) for quantification of plasma 25-OH vitamin D. Blood samples from 121 consecutive patients were collected and analyzed for the method comparison. Mean 25-OH vitamin D levels obtained by the HPLC method were found to be higher than the other two methods. However, statistically significant difference was found only with ECLIA (p=0.011). The three methods were compared by linear regression and the highest correlation was found for HPLC and ECLIA (r = 0.869), intermediate for CMIA-ECLIA (r = 0.818) and lowest for HPLC-CMIA (r = 0.779). As a result, the higher 25-OH vitamin D values were obtained with HPLC method, but the three methods showed good correlation and agreed well with each other. So that preferring the chemiluminescence methods which can be used in automated systems, seems to be advantageous.
