Analysis and Diagnosis of Mycovirus dsRNA in Verticillium dahliae, The Causal Agent of Verticillium Wilt Disease of Cotton

Abstract

Verticillium
dahliae

(Kleb), one of the important diseases that causes significant yield losses wherever
the cotton and olive were produced. Because the pathogen is soil-borne which
makes the management of the disease difficult. Therefore, biological control in
fungal diseases is becoming more essential, one of them is use the mycovirus
dsRNAs. Three studies on Verticillium
dahliae
mycoviral dsRNAs have been carried out so far, two of them in cotton
and one in olives. In Turkey, no studies were found on the presence of
mycoviral dsRNA in V. dahliae, which
causes cotton wilting. In this study, the presence of mycovirus dsRNA in 72 V. dahliae isolates obtained from the
cotton fields of Aydn/Söke and Koçarlı districts was investigated method. In 12
V. dahliae isolates dsRNA was
determined. The specific primers and random hexamer primer designed for the
mycovirus of V. dahliae (VdPV1-ol) in
olive were used in RT-PCR identification of this determined dsRNA. These primers
have been reported to have identical genome sequences with V. dahliae micovirus (VdPV1) in cotton. Furthermore, V. dahliae isolates containing dsRNA did
not respond to the apple pathogenicity test.

Pamuk Solgunluk Hastalık Etmeni Verticillium dahliae Kleb.’da Mikovirüs dsRNA Analizi ve RT-PCR İle Tanılanması

Abstract

Verticillium
dahliae
Kleb.
pamuk ve zeytin üretimi yapılan her yerde ciddi verim ve bitki kayıplarına
neden olan önemli bir fungal hastalıktır. Hastalığın toprak kökenli olması
mücadeleyi zorlaştırmaktadır. Fungal hastalıklarda biyolojik mücadele gün
geçtikçe ön plana çıkmakta olup biyolojik ajan olarak mikoviral dsRNA’lar da
kullanılmaya başlamıştır. Verticillium
dahliae
mikoviral dsRNA’lar ile ilgili ikisi pamukta biri zeytinde olmak
üzere toplamda dört çalışma yapılmıştır. Ancak Türkiye’de pamuk bitkisinde
solgunluk hastalığına neden olan V.
dahliae’
da mikovirüs dsRNA’nın varlığı hakkında bir çalışmaya
rastlanılmamıştır. Bu çalışmada Aydın ili Söke ve Koçarlı ilçelerinin pamuk
alanlarından solgunluk belirtisi gösteren 100 örnekten elde edilen 72 V. dahliae izolatında mikovirüs
dsRNA’nın varlığı araştırılmış ve on iki V.
dahliae
izolatında dsRNA belirlenmiştir. V.dahliae mikoviral dsRNA’nın RT-PCR ile tanılanması zeytindeki Verticillium dahliae partitivirus
1–olive (VdPV1-ol) mikovirüsü için tasarlanan spesifik primerler ve rastgele
hegzamer primeri kullanılarak yapılmıştır. Sonuçta kullandığımız bu primerler
ile pamuktaki V.dahliae izolatlarındaki
dsRNA mikovirüsünün tanılanamayacağı saptanmıştır. Çalışmada dsRNA içeren V. dahliae izolatlarının virülensliğini
belirlemek için elma testi uygulanmış ancak sonuç alınamamıştır.
