Catharantus roseus L.’den Elde Edilen Kallus ve Çoklu Sürgünlerde Alkaloidlerin Araştırılması

Abstract

Bu çalışmada içerdiği alkaloid sayısı bakımından zengin olan ve özellikle kanser tedavisinde büyük ilgi uyandıran Catharanthus roseus L. bitkisinde in vitro steril bitkiciklerin kök ve sürgün eksplantları ile kallus ve bir bitkicikten, çoklu sürgün üretimi araştırılmış olup, kalluslarda ve sürgünlerde alkaloid analizi yapılmıştır. C. roseus tohumları öncelikle Murashige-Skoog (MS) ve ½ MS besin ortamında çimlendirilmiş, 21 günlük bitkiciklerin kök ve sürgün kısmı ayrılmış ve iki farklı eksplant olarak (kök ve kotiledonlu hipokotil) farklı bitki büyüme düzenleyicileri ilaveli MS ortamında kallus üretimine teşvik edilmiştir. 0. 1 mg L-1 kinetin+ 1 mg L-1 NAA ve 0.1 mg L-1 kinetin+ 2 mg L-1 2.4 D ilaveli besin ortamlarında üretilen kallus ekstrelerinde triptofan alkaloidinin baskın olarak varlığı tespit edilmiştir. Ayrıca 1/2 MS ortamınında in vitro steril bitkicikler (21 günlük) köklerinden ayrılıp çoklu sürgün gelişimini teşvik için farklı konsantrayonlardaki BA ilaveli MS ortamında kültüre alınmıştır. Kontrol olarak in vivo koşullarda yetiştirilmiş olan (çiçeklenme döneminde temin edilmiş) C. roseus bitkilerinden alınan örneklerde (kök ve sürgün) alkaloid analizleri yapılmıştır. En iyi çoklu sürgün gelişimi 1 mg L-1 BA ilaveli besin ortamında görülmüştür. Bu besin ortamında yetişen sürgünlerden elde edilen alkaloidlerin kontrol bitkisi ile aynı bileşikler olmasının yanı sıra elde edilen serpentin indol alkaloidinin kontrol grubuna göre miktarı daha yüksek oranda bulunmuştur. Ayrıca triptamin alkaloidine de yalnızca kontrol ve 1 mg L-1 BA ilaveli besin ortamından elde edilen çoklu sürgünlerde rastlanılmıştır.

Investigation of Alkaloids in Callus and Multiple Shoots Obtained from Catharantus roseus L.

Abstract

In this study, the in vitro sterile plantlets and callus production in Catharanthus roseus L., which is rich in the number of alkaloid and arouses interest especially in cancer treatments, the multiple shoot production from sterile plantlets and the alkaloid analysis were made in both productions. C.roseus seeds were firstly germinated in full-power MS medium, the hypocotyl, cotyledon and root explants were promoted to callus production in different plant growth regulators supplemented basic MS medium. The dominant presence of a class of alkaloid (tryptophan) was identified in the 0. 1 mg L-1 kinetin+ 1 mg L-1 NAA and 0.1 mg L-1 kinetin+ 2 mg L-1 2.4 D supplemented nutrition environment. In the study, the plantlets (root and hypocotyl + cotyledon) germinated in 1/2 MS medium were also separated from their roots and were cultivated in basic MS medium supplemented with different concentrations of BA for promotion of multiple shoot growth. To control alkaloid analyses were made for in vivo grown C.roseus plant (provided during the flowering period). The best shoot growth was seen in 1 mg L-1 BA supplemented nutrition environment and the obtained serpentine indol alkaloid was determined to be higher than the control plant although alkaloids obtained from the shoots grown in that nutrition environment were the same compounds as the control plant. Additionally, tryptamine alkaloid occurred only in control and the shoots obtained from that nutrition environment.
