BIOCHEMICAL EVALUATION OF DEXPANTHENOL ON INTESTINAL ISCHEMIA-REPERFUSION INJURY IN RATS

Öz

Objective: This study investigated the protective and treated effects of dexpanthenol (DEX) on intestinal injury caused by ischemia-reperfusion (I/R). Methods: Seven groups of rats were used: a control group, a sham-operated group, an I/R group, two groups receiving DEX before ischemia (I/R+Pre250 mg/kg and I/R+Pre500 mg/kg), and two groups receiving DEX after ischemia (I/R+Post250 mg/kg and I/R+Post500 mg/kg). Ischemia was induced by clamping the superior mesenteric artery for 45 minutes, followed by 120 minutes of reperfusion. Intestinal tissue samples were collected to assess enzyme levels. Results: The induction of I/R injury increased the levels of malondialdehyde, myeloperoxidase, catalase, and glutathione peroxidase. DEX treatment especially increased glutathione levels in the ileum, the most effective treatment regimen was observed in the I/R+Post500 group. This dose is also limited to increment of malondialdehyde, myeloperoxidase, nitric oxide and catalase levels. Glutathione reductase levels were not altered by either I/R injury induction or DEX treatment. Conclusion: Intravenous administration of 500 mg/kg DEX attenuated intestinal ischemia-reperfusion injury in rats, particularly when administered immediately after the onset of reperfusion. The oxidant and antioxidant modulating effect of DEX was dose-dependent.
