Detection of the Activation of Nuclear Factor Kappa Βeta NF-KB Signalization Pathway in Peripheral Giant Cell Granulomas

Öz

The aim of this study is to detect the activity of NF-KB signalization system in peripheral giant cell granulomas in order to determine the origin of the pathogenesis of the lesion. The clinical data and paraffin blocks of twenty nine cases of peripheral giant cell granuloma were retrieved from the archives. Glass slides were prepared from retrieved paraffin blocks and subsequently immunostained with NF-KB antibody. 97 % of the selected cases were highly or moderadetly stained with NF-KB immunostaining. All cases showed reactivity with NF-KB immunostain. The detection of the activation of NF-KB pathway in peripheral giant cell granulomas suggest that these lesions may have an inflammatory origin. In future studies, modification of NF-KB pathway with drugs may reduce the growth potential or prevent the occurrence of the peripheral giant cell granuloma

Periferal Dev Hücreli Granülomalarda Nükleer Faktör Kappa Beta NF-KB Sinyal Yolağının Aktivitesinin Araştırılması

Öz

İnflamatuar gen anlatımında çok büyük rolü olan nükleer faktör kappa beta NF-KB sinyal sisteminin periferal dev hücreli granülomalarda aktif olup olmadığını araştırmak, böylece bu lezyonların oluşumlarının inflamatuar bir patogenezden köken alıp almadığı konusunda fikir sahibi olabilmektir. Retrospektif olarak elde edilmiş 29 vakanın klinik bilgileri incelenip, parafin bloklardan elde edilen yeni kesitler üzerinde immunohistokimyasal NF-KB boyaması yapıldı. Tüm vakaların % 97’sinde şiddetli ve orta şiddetli boyanma tespit edildi. Bütün vakalarda NF-KB aktivasyonu gözlendi. NF-KB sinyal yolağının periferal dev hücreli lezyonlarda aktif olması bu lezyonların inflamatuar kökenli patolojiler olduğunu göstermektedir. Gelecek çalışmalarda NF-KB sinyal sisteminin ilaçlarla modifiye edilmesi ile bu lezyonlara karşı durdurucu tedavi stratejileri geliştirilebilir
