The determination of Parainfluenza-3 Viral Antigens Using Immunohistochemical Methods in Cattle Lungs in Erzurum Region

Öz

The aim of this study was to investigate immunohistochemically the presence and the prevalence of Parainfluenza-

3 (PI-3) in the lungs of cattle breeding in the Erzurum region. For this aim, the lung samples of 600 cattle slaughtered in

abattoirs in Erzurum province were examined by collecting and pneumonia was macroscopically observed in 75 (12.5%) lung

samples. 58 interstitial pneumonia, 10 broncho-interstitial pneumonia, 2 catarrhal pneumonia, 2 fibrinous pneumonia, and 3

granulomatous pneumonia were observed in the histopathological evaluation of these lungs examined macroscopically.

Immunohistochemical staining was performed to pneumonia detected lung sections in terms of PI-3 virus and

immunopositivity was detected in 7 (9.33%) lung samples. Immunopositivity was observed in 6 (10.34%) interstitial

pneumonia and 1 (10%) broncho-interstitial pneumonia in lung samples. Immunopositivity in the lungs which were positive

for PI-3 viral antigens was found in the cytoplasm of bronchi and bronchioles epithelium, in inflammation cells in bronchusassociated

lymphoid tissue (BALT) around bronchioles, in the alveoli, in the macrophages in the interstitial area. Furthermore,

these positivities were observed in the inflammatory exudate in the bronchial lumen in the broncho-interstitial pneumonia

lung samples. In conclusion, it was determined that the presence of PI-3 viral antigens has an important place in agents which

occur pneumonia in cattle in Erzurum region and it is thought that this agent should not be ignored in the protection and

treatment methods against the bovine respiratory disease complex.

Erzurum Yöresinde Sığır Akciğerlerinde Parainfluenza-3 Viral Antijenlerinin İmmunohistokimyasal Yöntemlerle Belirlenmesi

Öz

Bu çalışmada, Erzurum yöresinde yetiştirilen sığırların akciğerlerinde Parainfluenza-3 (PI-3) etkeninin immunohistokimyasal olarak varlığının ve yaygınlığının araştırılması amaçlanmıştır. Bu amaçla, Erzurum ilinde faaliyet gösteren kesimhanelerde kesimi yapılan 600 sığıra ait akciğer örneği toplanarak incelendi ve makroskobik olarak 75 (%12.5) akciğer örneğinde pnömoni bulgusuna rastlandı. Makroskobik olarak incelenen bu akciğerlerin histopatolojik incelemesinde 58 intersitisyel pnömoni, 10 bronkointersitisyel pnömoni, 2 kataral bronkopnömoni, 2 fibrinli pnömoni ve 3 granulomatöz pnömoni olgusu gözlendi. Pnömoni saptanan akciğer kesitlerine PI- 3 virusu yönünden immunohistokimyasal boyama yapıldı ve 7 (%9.33) akciğer örneğinde immunpozitiflikler belirlendi. İmmünpozitifliklere intersitisyel pnömoni gözlenen 6 (%10.34), bronkointersitisyel pnömoni gözlenen 1 (%10) akciğer örneğinde rastlandı. PI-3 viral antijeni pozitif olan akciğerlerde immunpozitifliklere bronş ve bronşiyol epitellerinin sitoplazmasında, bronşiyol çevresindeki bronşiyoalveolar lenfoid dokudaki (BALT) yangı hücrelerinde, alveollerde, intersitisyel alandaki makrofajlarda rastlandı. Ayrıca bu pozitiflikler bronkointersitisyel pnömoni gözlenen akciğer örneklerinde bronşiyol lümenindeki yangısal eksudatta gözlendi. Sonuç olarak; Erzurum yöresindeki sığırlarda pnömoni oluşturan etkenler içerisinde PI-3 viral antijen varlığının önemli bir yere sahip olduğu belirlenmiş ve sığırlardaki solunum sistemi hastalık kompleksine karşı yapılan koruma ve tedavi yöntemlerinde bu etkenin göz ardı edilmemesi gerektiği düşünülmüştür.
