Investigation of the Effect of Extraction Temperature on Phenolic Acids and Flavonoids in Matricaria chamomilla L. Using Accelerated Solvent Extraction

Matricaria chamomilla L. is a commonly utilized medicinal plant abundant in phenolic compounds that enhance its biological and therapeutic attributes. The efficacy and specificity of phenolic extraction are significantly affected by extraction parameters, especially temperature. This work utilized accelerated solvent extraction (ASE) to examine the influence of extraction temperature on the phenolic composition of Matricaria chamomilla extracts. Extractions were conducted utilizing methanol at five distinct temperatures (40, 50, 60, 80 and 100 °C) under consistent pressure and extraction conditions. The resultant extracts were examined via UHPLC–MS/MS for the quantitative assessment of phenolic acids and flavonoids. The findings demonstrated a distinct temperature-dependent pattern, with the peak concentrations of the majority of phenolic chemicals achieved at 40 °C. Elevating extraction temperature resulted in a significant reduction of both phenolic acids and flavonoids, suggesting thermal sensitivity and potential destruction at higher temperatures. Minor recoveries for specific chemicals were noted at 100 °C; however, these effects were constrained. The findings indicate that mild ASE conditions are superior for maintaining phenolic integrity in chamomile extracts and emphasize the necessity of temperature adjustment in ASE-based phytochemical extraction.
