Synthesis of Organophilic Clay-Nanocomposite Containing Sumac Extract for Dermatological Purposes and Investigation of Its Bioactivity

Abstract

The use of clays for cosmetic purposes has existed since ancient times, and its therapeutic use has existed since prehistory, and today's cosmetic industry uses significant amounts worldwide. Antioxidant and antibacterial compounds are generally used as active ingredients in many cosmetic products. In this study, it was aimed to prepare bioactive organophilic nanocomposites with an innovative approach in order to increase the effectiveness of clay (montmorillonite, MMT) mineral, which is widely used in cosmetic formulations, in line with its intended use and to test its bioactivity. For this purpose, montmorillonite clay obtained from domestic sources was purified by sedimentation method. Montmorillonite clay was made organophilic (OMMT) by using dodecyltrimethyl ammonium chloride solution. Sumac (fruit part), which is among the medicinal plants and used as a spice, was taken from the attar, dried, ground and extracted with water, ethanol and n-hexane separately for 8 hours in a Soxlet extraction apparatus. The n-hexane in the n-hexane extract was removed under vacuum and the obtained solid was extracted again with water/ethanol solution to obtain food grade sumac extract with 95% yield. The solvents of water and ethanolic extracts were also removed under vacuum. Nanocomposites were prepared with 1%, 3% and 5% sumac extract amount in OMMT and their bioactivity was tested by compatibilization with PEG-400. Bioactivity tests were performed on Nutrient Agar and Violet Bile Red Agar media at 37oC using gram positive Staphylococcus aureus and gram negative Escherichia coli bacteria growing on the skin surface. All of the clay nanocomposites prepared in three different compositions showed that they had bioactive properties by inhibiting the growth of Staphylococcus aureus and Escherichia coli bacteria.
