The Effects of Ginkgo Biloba Extracts on the Nephropathy of Diabetic Rats Induced By Streptozotocine (STZ)

This study was designed to evaluate the effects of Ginkgo biloba extracts on the kidneys of diabetic rats induced by streptozotocine (STZ). 40 adult male Wistar albino rats were examined. Rats were divided into four groups as control, diabetic group, Ginkgo biloba administered diabetic group and group exposed to solvent only. Experimental diabetes was induced by intraperitoneal application of 60 mg/kg STZ to diabetes group and Ginkgo biloba administered diabetes group. After the induction of diabetes, every day 100 mg/kg Ginkgo biloba extract was administered orally to the ginkgo group for 40 days. Throughout the experiment blood glucose levels and body masses of the rats was monitored. On the 40th day rats were decapitated under ether anesthesia and their kidneys were removed. The tissue samples were embedded in paraffine. After histochemical staining the sections were investigated under lightmicroscopy. While blood glucose level of the diabetes group was significantly higher compared with the control and solvent groups, the body mass significantly decreased. Basic glomerular and tubular histologic changes caused by diabetes, like thickening of the basal membrane, expansion of the mesenchymal matrix, glomerulosclerosis, tubular necrosis, hydropic degeneration, cellular swelling, vacuolization, were observed. These changes were observed significantly less in the Ginkgo biloba group Gingko biloba extract (Egb 761) decreased kidney damage in diabetic rats induced with STZ. We therefore believe that Ginkgo biloba extracts may help preventing the development of kidney damage and decrease the amount of pathologic findings in diabetes. Further studies are still necessary to demonstrate the efficacy of long term Ginkgo biloba use on diabetic complications.

Streptozotosinle İndüklenen Diabetik Ratlarda Ginkgo Biloba (EGb 761) Ekstresinin Nefropati Üzerine Etkileri

Bu çalışma, streptozotosin (STZ) ile indüklenen diabetik ratların, böbreklerinde oluşan histolojik değişiklikler üzerine ginkgo bilobanın etkilerinin araştırılması amacıyla planlandı. Çalışmada Wistar Albino cinsi 40 adet erişkin erkek rat kullanıldı. Ratlar, kontrol, diabetik grup, Ginkgo verilen diabetik grup ve çözücü verilen grup olmak üzere 4 gruba ayrıldı. Deneysel Diabet, diabetik ve Ginkgo biloba verilen diabetik gruplarda tek doz STZ'nin (60 mg/kg) intraperitoneal uygulanmasıyla oluşturuldu. Diabet oluştuktan sonra, Ginkgo biloba grubuna 40 gün süresince her gün 100 mg/kg ginkgo biloba ekstresi oral olarak verildi. Deney süresince ratların kan-glikoz seviyeleri ve vücut ağırlıkları takip edildi. 40. gün ratlar eter anestezisi altında dekapite edilerek böbrekler alındı. Dokular rutin takip sonrası, parafine gömüldü. Histokimyasal boyama sonrası, kesitler ışık mikroskopta incelendi. Diabet grubundaki ve Ginkgo verilen gruptaki ratların, kontrol ve çözücü verilen gruba göre kan- glikoz seviyeleri önemli oranda artarken, vücut ağırlıklarıda önemli oranda azaldı. Diabetik grupta glomerüllerde mezengial matriks genişlemesi bazal membran kalınlaşması, glomerüloskleroz, tübüllerde vakuolizasyon, tübüler nekroz, hidropik dejenerasyon ve hücre şişmesi gibi temel histolojik değişiklikler gözlendi. Uygulanan Ginkgo biloba ekstresi ile bu bulguların önemli derecede azaldığı tespit edildi. Ginkgo biloba ekstresi (EGb 761), Streptozotosinle ratlarda oluşturulan diabetin sebep olduğu böbrek hasarlarını azalttı. Bu yüzden Ginkgo biloba ekstresinin diabetik böbrekte görülen patolojik bulguları azaltacağını ya da gelişimini önleyebileceğini düşünmekteyiz. Yinede diabetik komplikasyonlar üzerindeki yararlı etkisini gösterebilmek için uzun süre kullanımıyla ilgili, ileri düzeyde çalışmaların yapılması gerekmektedir.
