Sığırlarda Swim-Up Yöntemi ile Sperma Separasyonunun Embriyo Cinsiyet Oranına Etkisi

Öz

Sunulan çalışmada sığır spermasında uygulanan farklı swim-up inkübasyon sürelerinin in vitro üretilen embriyolarda cinsiyet oranına etkisi araştırılmıştır. İn vitro mature edilen oositler üç gruba ayrıldı. Birinci, ikinci ve üçüncü gruptaki mature oositler sırasıyla 5, 10 ve 15 dakika süre ile swim-up işlemi yapılan spermalar ile fertilize edildi. Altmış dakika süre ile inkübasyon yapılan spermalarla yapılan tohumlamadan elde edilen embriyolar kontrol grubu olarak kullanıldı. Her bir gruptan elde edilen 10 embriyoda polimeraz zincir reaksiyonu (PZR) yöntemi kullanılarak cinsiyet (erkek oranı) oranı belirlendi. İlk grupta hiç erkek embriyo elde edilemedi. İkinci grupta bir adet erkek embriyo elde edildi. Üçüncü ve kontrol gruplarında ise iki adet erkek embriyo elde edildi. Sonuç olarak bu çalışma sonunda in vitro embriyo üretiminde farklı sürelerde swim-up yapılmasının sığırlarda yavru cinsiyet oranı üzerine etkisi bulunamamıştır. Daha doğru sonuçların alınması için yeni in vivo ve in vitro çalışmalar gerekmektedir
