RATLARDA TİYOSEMİKARBAZON TÜREVLERİNİN BAZI KAN PARAMETRELERİNE ETKİLERİ

Öz

Çalışmada, deney grubu ratlara 45 günlük deneysüresince üçer gün ara ile deri altı yolla 25 mg/kgtiyosemikarbazon ve 25 mg/kg tiyosemikarbazon-çinkokompleksi uygulandı. Uygulamaların serum aspartatamino transferaz (AST), alanin amino transferaz (ALT),laktat dehidrogenaz (LDH), gamma-glutamil transferaz(GGT) aktivitelerine ve üre, kreatinin, total ve direktbilirubin düzeylerine, karaciğer ağırlığına ve canlıağırlığına etkileri belirlemek için 60 adet rat kullanıldı.Ratlar kontrol grubu, tiyosemikarbazon uygulanan grupve tiyosemikarbazon-çinko kompleksi uygulanan deneygrubu olmak üzere üç gruba ayrıldı. Uygulamanın 7.,15., 30. ve 45. günlerinde her gruptan 5’er rat ayrılıptartılarak dekapite edildi; kanları alınıp karaciğerağırlıkları kaydedildi. Gerekli analizler otoanalizördespektrofotometrik yöntemle yapıldı.Tiyosemikarbazon ve tiyosemikarbazon-çinko kompleksiuygulanan ratların serum AST, ALT, LDH ve GGTaktiviteleri ile üre, kreatinin, total ve direkt billirubindüzeyleri, ağırlıklarının kontrol grubuna göre istatistiksel olarakönemli bir fark göstermediği (p>0.05) bulundu. Ratlaraderi altı yolla verilen 25 mg/kg tiyosemikarbazon vetiyosemikarbazon-çinko kompleksinin çalışılan serumparametrelerini, karaciğer ağırlığını ve canlı ağırlığıetkilemediği gözlendi.Buna tiyosemikarbazon-çinko kompleksinin tedavi amaçlıkullanıldığında etkilerinden korunulabileceği düşünüldü

The Effect of Thiosemicarbazone Derivatives on Some Blood Parameters in Rats

Öz

In this study, 25 mg/kg thiosemicarbazoneand 25 mg/kg thiosemicarbazone-zinc complex weregiven to the experiment group rats subcutoneusly at aninterval of three days. 60 rats were used to determine ofthe effect of thiosemicarbazone and thiosemicarbazonezinc complex on the rats, the activities of aspartateaminotransferase (AST), alanine aminotransferase(ALT), lactate dehydrogenase (LDH), gamma-glutamyltransferase (GGT) and levels of urea, creatinine, directbillirubin, total billirubin in serum and liver weight andlive weight of rats.Rats were divided into three groups namely the controlgroup and the two experimental groups exposed tothiosemicarbazone complex.Five rats from the each group at 7th, 15th, 30th, 45thdaysof application were separated and weighed and thendecapitated. Their bloods were taken and their liverweights were recorded. Necessary analyses wereconducted through the autoanalyser and no statisticallysignificant differences were found in activities of AST,ALT, LDH, GGT, and levels of urea, creatinine, totaland direct billirubin in serum of rats exposed tothiosemicarbazone complex, and their liver weight and alive weightsrelative to control group (p>0.05).It has been observed that 25 mg/kg thiosemicarbazoneand 25 mg/kg thiosemicarbazone-zinc complex givensubcutoneusly to the rats did not affect some of theirblood parameters, and liver and live weights of rats.As a result, it was thought that when thiosemicarbazoneand thiosemicarbazone-zinc complex were used for thepurpose of treatment, from their side effects might beavoided by adjusting suitable doses of those substances
