Büyük Hacimde Süspanse Hücre Kültürlerinin Stoklanması Üzerinde Uygulamalar

Öz

Büyük hacimdaki hücrelerin uzun süreli muhafazası için -130·c nin altındaki sıcaklıklara ihtiyaç vardır. -196.C'ye dayanıklı hücre dondurma ampullerinin maksimum kapasitesi 5 ml'dir. 100 ml hacimdaki hücrelerin dondurularak muhafazası için ticarette sadece palikarbonat ampuller bulunmaktadır ki bunlar da ancak -150·c·ye kadar bozulmadan kalabilmektedirler. Bu nedenle denemeler sıvı azot gazında hücre muhafazası konusunda yoğunlaştırılmıştır. Bu çalışmada 15 seri hücre kültürü 100 ml hacimda dondurulmuştur. Bu hücrelerin çözdürüldükten sonraki üreyebilirlik oranının ortalama % 90 olduğu tespit edilmiştir.

A Study on the stocking of the suspended cell oultiıres in lorge volumes

Öz

For long term storage of so large quantities of cells it, ls necessary to keep them below minus 130·c. The maximum capacity of the ampoules which are not demaged in minus 196·c is limited with 5 ml' To freeze the cells in at least 100 ml volume only polycarbonate ampaules are commercially available. Those are resistant to steam sterilisation and freezing untill minus 156·c. For that reason the experiment is realised in the gas phase of liquid nitrogen.
