Borrel Metodu ile Üretilen Koyun Çiçek Virusunun Koyunlarda ve Doku Kültürlerinde Mukayeseli Olarak Titrasyon Testlerinin Saptanması

Comparative titration test of sheep pox virus produced by Borrel method on sheep and sheep kidney tissue culture.

Öz

This study was conducted to compare th e titration of sheep
pox virus produced by Borrel method on sheep and sheep kidney
tissue culture. 8 dilutions of the virus with 1/10.000 to
1/1.280.000 were used for titrations.
Results in sheep were appaered acrording to the pustules
and dose minimal reaction (DMR) after intadermicly injections
of the vaccine virus dilutions. Any sheep refractory in the
exper~ment were disc?trded. 21 .sheep were used in the study and
they were kıvırcık race which were more susceptible to the
virus.
Results in the sheep kidney tissue culture were fallowed
· for cytopathic efect (CPE) under microscope for 12 days. CPE
were appaered by the method of Reed and Muench and explained
by tissue culture infective dose (TCID/50).
· Titrations on sheep were estımated as being with a range
of 10-5,80 - 10-6,40 where as an tissue culture 10-5,65 10-6,35 the results of the both virus titrations were similar to
each other, but titrations of virus on sheep kidney tissue
culture were obviosly more economic and practical than the
regular method with pustules on sheep.
