Detection and molecular characterization of Genogroup I Picobirnaviruses in dogs

Öz

Picobirnaviruses (PBVs) have been reported in other terrestrial and marine mammals with or without diarrhea, birds, invertebrates, as well as environmental aquatic samples with diarrhea or asymptomatic since they were first detected in stool samples from humans and rats in 1988. However, in dogs, there are only a few reports of PBV detection and its molecular epidemiology. In this study, it was aimed to detect and molecular characterization of PBVs in a total of 75 stool samples obtained from puppies aged 0-6 months with diarrhea and clinically healthy. For this purpose, the RT-PCR was performed by using primers targeting the RdRp gene of genogroup I (GGI) PBV, and four of the tested samples (5.33%) were found positive for GGI PBV. One of these samples (CB1) was obtained from a dog with diarrhea, and the other three samples (KB19, KB29, KB30) were obtained from clinically healthy dogs. In this study, the presence and molecular characterization of PBV in dogs with diarrhea and clinically healthy appearance were revealed for the first time in our country.

Sağlıklı ve ishalli köpeklerde Genogrup I Picobirnavirusların tespiti ve moleküler karakterizasyonu

Öz

Picobirnaviruslar (PBV) ilk olarak 1988'de insan ve sıçanların dışkı örneklerinde tespit edilmelerinden bu yana ishalli ya da asemptomatik diğer kara ve deniz memelilerinde, kuşlarda, omurgasızlarda ve ayrıca çevresel su örneklerinde rapor edilmiştir. Buna karşın, köpeklerde, PBV tespiti ve moleküler epidemiyolojisi hakkında sadece birkaç rapor vardır. Bu çalışmada, klinik olarak ishal semptomu olan ve sağlıklı görünen 0-6 ay yaş arasındaki yavru köpeklere ait toplam 75 adet dışkı örneğinde PBV’lerin tespiti ve moleküler karakterizasyonu hedeflenmiştir. Bu amaçla örneklere, genogrup I (GGI) PBV’nin RdRp genini hedefleyen primeler kullanılarak RT-PCR uygulanmış ve test edilen örneklerin dört tanesi (%5.33) GGI PBV yönünden pozitif bulunmuştur. Bu örneklerden biri (CB1) ishalli bir köpekten, diğer üç örnek (KB19, KB29, KB30) ise klinik olarak sağlıklı görünümlü köpeklerden elde edilmiştir. Bu çalışma ülkemizde ilk defa ishalli ve klinik olarak sağlıklı görünümlü köpeklerde PBV varlığı ve moleküler karakterizasyonu ortaya konulmuştur.
