Monoklanal Antikorların Newcastle Hastalığının Teşhis ve Epidemiyolojisinde Kullanımı

Öz

Poliklonal antikorların konvansiyonel serolajik testlerde virusların
ayrımında kullanılması bazı problemlere yol açmaktadır. Monoklonal
antikor teknolojisi (mAb) konu için yeni bir alternatif olmuştur.
Newcastle Hastalığı Viruslarına (NHVJ karşı hazırlanan mAb'ler
değişik izolatlar arasındaki kayda değer antijenik farklılığı göstermişlerdir.
NHV suşlarının hepsi yada pek çoğu için spesifik olan ve HI aktivitesi
gösteren mAb'ler. Newcastle Hastalığı (NH) için referans laboratuvar
olan Weybridge'de rutin teşhiste kullanılmaktadır. Bu
mAb'lerin kullanımı NHV. variant NHV. Lasota ve PMV-3 arasında ayrımı
sağlamaktadır.
MAb'ler aynı zamanda NHV'larının antijenik yapılarına göre
gruplanmalarını sağlamışlardır. Bu konudaki en başarılı yaklaşım
Russell ve Alexander ' ın çalışmaları olmuştur. Araştırıcılar, NHV'larını,
NHV Ulster 2C suşuna karşı oluşturulan 9 mAb ile reaksiyonlarını
temel alarak gruplamışlardır. Oluşturulan grupların virulans, temel
konakçı gibi epidemiyolojikal özellikleri büyük ölçüde paylaştığı
görülmüştür. MAb gruplamasının NH'nin epidemiyolojisindeki başarısı.
İngiltere'de variant NHV'larının tavuklarda oluşturduğu NH salgınlarını
ve Türkiye'de Güney Marmara bölgesinde son zamanlarda
görülen NH salgınını da içeren pek çok olguda kanıtlanmıştır.

Use of Monoclonal Antibiotics in Diagnosis And Epidemiology or Newcastle Disease

Öz

Applications of monoclonal an tibody (mAb) technology for Virus
characterisation has been an alternative approach since polyclonal
antibodies have been shown to have some problemsin discriminating
Viruses in conventional serological tests . MAbs to Newcastle disease
virus (NDVJ have proved the considerable variation among different
isola tes. Many laboratories have prepared mAbs to NDVs and shown
conserved and variant epitopes on some isolates.
MAbs that is spesific for all or vast majority of NDV strains and
show HI actiVities are now being used in Weybridge. international referans
laboratory for Newcaslle Disease (ND), for routine diagnosis.
Use of these mAb's enables distinction between NDV, variant NDV.
Lasota and PMV-3 Viruses.
MAbs also served to group NDVs with regard to their antigenic
properties. The most successful approaches on this subject were studies
of Russell and Alexander. They grouped NDV strains on the hasis
of their reaction with 9 mAbs to NDV Ulster 2C. The groups formed
have shared im portant epidemiological properties, such as main
host, virulance ete. Success of mAb grouping in epidemiology of the
ND has been confirmed in many ND casesincluding ND outbreakin
chickens due to variant NDVs in England and recent ND outbreakin
south Marmara region of Turkey.
