Adenozin Deaminaz Enziminin Kinetik Davranışı Üzerine Zerdeçal (Curcuma Longa) Ekstresinin Etkilerinin Araştırılması

Öz

Bu çalışmada, kanser metabolizmasının kilit enzimlerinden biri olan adenozin deaminaz (ADA) sığır karaciğerinden izole edilerek kısmen saflaştırılmış, kinetik özellikleri incelenmiş ve enzimin kinetik davranışı üzerine zerdeçal ekstresinin etkileri araştırılmıştır. ADA enziminin inhibitörsüz şartlarda çizilen Michaelis Menten ve Lineweaver Burk grafiklerinde V max değeri absorbans olarak 0,8 (spesifik aktivite olarak 40 mIU/g protein), Km değeri 100 mM olarak tespit edilmiştir. Zerdeçal ekstresinin iki farklı konsantrasyonunda çizilen Michaelis Menten ve Lineweaver Burk grafiklerinde ise her iki durum için Km değeri 100 mM olarak, V max değerleri ise % 10 zerdeçal konsantrasyonunda 0,5 (25 mIU/g protein), % 20 zerdeçal konsantrasyonunda ise 0,3 (15 mIU/g protein) olarak bulunmuştur. Bu sonuçlar zerdeçal bileşenlerinin ADA enzimini non-kompetetif türde inhibe ettiğini göstermektedir. ADA enziminin zerdeçal tarafından bu şekilde inhibisyonu, zerdeçalın anti kanser özelliklerine katkı sağlayan mekanizmalardan biri olabilir.
