Yaşlı Sıçanlarda Antrenman ve Koenzim Q10 Takviyesinin Kalp Dokusunda Oksidatif Stres ve Antioksidan Savunma Belirteçleri Üzerine Etkileri

Öz

Amaç: Bu çalışmanın amacı, yaşlı sıçanlarda antrenman ve/veya koenzim Q10 CoQ10 desteğinin kalp dokusunda oksidatif stres ve antioksidan savunma değişiklikleri üzerine etkilerini incelemekti. Gereç ve yöntem: Çalışmada 27 genç 3 aylık ve 27 yaşlı 20 aylık Wistar erkek sıçan kullanıldı. Her bir grup; kontrol, antrenman, CoQ10 ve CoQ10+antrenman olmak üzere dörder alt gruba ayrıldı. Antrenman gruplarındaki sıçanlara 8 hafta boyunca, haftada 5 gün, günde 1 saat yüzme egzersizi yaptırıldı. CoQ10 desteği alan sıçanlara 8 hafta boyunca günde 300 mg/kg CoQ10 oral olarak verildi. Sıçanlar, anestezi altında kalp dokusu örnekleri alındıktan sonra servikal dislokasyonla sakrifiye edildi. Alınan örneklerde malondialdehit MDA , 8-hidroksi–2-deoksiguanozin 8-OHdG , glutatyon GSH seviyeleri ve süperoksit dismutaz SOD ve katalaz CAT aktiviteleri ölçüldü. Bulgular: Gruplar arasında MDA, 8-OHdG ve GSH seviyelerinde herhangi bir fark yoktu. SOD ve CAT aktivitesi genç CoQ10 ve CoQ10+antrenman gruplarında ve tüm yaşlı gruplarda, genç kontrol ve genç antrenman gruplarından düşüktü. Sonuç: Yaşlanma, CoQ10 takviyesi ve antrenman, kalp dokusunda lipid peroksidasyonu, DNA hasarı ve enzimatik olmayan antioksidan seviyeleri üzerine sınırlı etkiye sahipken, antioksidan enzim aktivitesini azaltır

Öz

Objectives: The aim of this study was to investigate the effects of exercise training and/or coenzyme Q10 CoQ10 supplementa- tion on oxidative stress and antioxidant defense changes in heart tissue of aged rats. Material and methods: Twenty-seven young 3-months of age and 27 aged 20-months of age Wistar male rats were used in the study. Each group was further divided into four subgroups: Control, training, CoQ10 and CoQ10+training. The training program consisted of swimming exercise one hour per day, five days a week, for eight weeks. The CoQ10 were given at a daily dose of 300 mg.kg-1 of body weight five days/week via oral gavage. Heart tissues were taken under anesthesia then rats were sacrificed by servical dislocation. Malondialdehyde MDA , 8-hydroxy-2-de- oxyguanosine 8-OHdG , glutathione GSH levels and superoxide dismutase SOD and catalase CAT activities were analyzed in samples.Results: There was no significant difference among the groups with respect to MDA, 8-OHdG and GSH levels. SOD and CAT activities were lower in the young CoQ10 and in the CoQ10 plus training groups than the young control, young training and all aged groups. Conclusion: Although aging, CoQ10 supplementation and exercise training have limited effect on lipid peroxidation, DNA damage and non-enzymatic antioxidant levels, they decreased antioxidant enzyme activity in the heart tissue
