E vitamini ve selenyumun deneysel diyabette trombosit ve eritrosit indeksleri ile pankreas histopatolojisi üzerine etkisi Türkçe

Öz

Amaç: Bu çalışmada, deneysel diyabette E vitamini ve Selenyum Se 'un eritrosit ve trombosit indeksleri ile pankreas dokusu üzerinde olası etkileri araştırılmıştır. Materyal ve metot: Çalışma beş gruba ayrılan Wistar albino ratlar üzerinde yürütüldü. Birinci gruptaki ratlar n=5 kontrol olarak kullanıldı, bunlara sadece yem ve su verildi. İkinci gruptaki ratlarda n=5 65 mg/kg 0,1 M sitrat tamponda hazırlanmış streptozotosin STZ intraperitoneal İP uygulanarak deneysel diyabet oluşturuldu. STZ enjeksiyonuna ilaveten, üçüncü gruba n=10 E vitamini alfa-tokoferol asetat, 100 mg/kg , dördüncü gruba n=10 Se sodyum selenat 0.3 mg/kg , beşinci gruba n=10 ise Se-E vitamini kombinasyonu sodyum selenat 0.3 mg/kg+alfa-tokoferol asetat, 100 mg/kg gün aşırı uygulandı. Onbeş gün süren çalışma sonunda tüm ratlardan kan alınarak hematolojik değerler belirlendi. Pankreastan alınan doku kesitleri histopatolojik olarak incelendi. Bulgular: Tüm gruplarda alyuvar sayısı, hemoglobin miktarı, hematokrit değer ile alyuvar hacmi, ortalama alyuvar hemoglobini, ortalama alyuvar hemoglobin yoğunluğu, alyuvar dağılım genişliği gibi alyuvar indeksleri; trombosit sayısı, ortalama trombosit hacmi, trombokrit/plateletkrit değeri ve trombosit dağılım genişliği gibi trombosit indeksleri ile akyuvar sayısı, nötrofil, lenfosit, monosit, eozinofil, bazofil sayıları ve bu lökositlerin oranları % otomatik cell-counter'de belirlendi. Sonuç: Se ve E vitamininin deneysel diyabette trombosit ve trombokrit/plateletkrit değerleri ile nötrofil, eosinofil, bazofil sayıları üzerinde etkili olduğu saptandı. Ancak diğer kan parametreleri üzerindeki etkilerinin istatistiksel anlamda önemli olmadığı, kontrol grubuna göre deneme gruplarında ışık mikroskobisi düzeyinde pankreas kesitlerinde önemli bir değişiklik oluşmadığı gözlemlendi

Effects of vitamin E and selenium on erythrocyte and platelet indices and pancreatic histopathology in experimental diabetes

Öz

Background: In this study, possible effects of Se and vitamin E were investigated on histopathology of pancreas, platelet and erythrocyte indices in experimental diabetes.Methods: All Wistar albino rats were randomly divided into five groups. The first group n=5 was used as a control and this group was received only food and water. The second group n=5 was used as diabetic control group and it was only made diabetic by intraperitoneal injection in one time with 65 mg/kg of streptozotosin STZ in citrate buffer. The third group n=10 received with Se Na SeO , 0.3 mg/kg body weight , the fourth group n=10 with vitamin E dl--tocopheryl acetate, 100 mg/kg body weight , and the fifth group n=10 with Se plus vitamin E combination Na SeO , 0.3 mg/kg body weight+dl--tocopheryl acetate, 100 mg/kg body weight for every other day intervals during fifteen days. Blood parameters of the all rats were analyzed in 15 days after STZ injection. Then, their pancreatic tissues were histopathologically examined.Results: Platelet indices such as platelet counts, mean platelet volume, plateletcrit and platelet distribution width, erythrocyte indices such as red blood cell conunts, hemoglobin, heamatocrit%, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, red cell distribution width and also, the values of white blood cell, neutrophil, lymphocyte, monocyte, eosinophil and basophil and percentage of these leukocytes % in blood samples of all groups were determined by automotic cellcounter. Conclusions: As a result, it was determined that Se and vitamin E affected the values of plateletcrit neutrophil, eosinophil and hematological parameters of these antioxidants. In the histopathologic results, we couldn't see any change or important effect on the pancreas tissue on the light microscope in all experimental groups.platelet and basophil There were not statistical changes on the values of other
