Development of Pink Tomato Breeding Materials Resistant to TYLCV and FORL Pathogens

Abstract Tomato Yellow Leaf Curl Virus (TYLCV) and Fusarium oxysporum f.sp. radicis-lycopersici (FORL) are among the most destructive pathogens limiting tomato (Solanum lycopersicum L.) production worldwide, causing substantial yield and quality losses. The development of cultivars with durable and multiple disease resistance has therefore become a major objective of modern tomato breeding. In this study, molecular markers were used to identify resistance genes against TYLCV and FORL in pink tomato breeding lines developed within the Istanbul Tarım A.Ş. tomato breeding program. Breeding materials previously characterized for resistance to root-knot nematodes (Meloidogyne spp.) and Tomato Spotted Wilt Virus (TSWV), together with additional breeding lines, were evaluated to identify genotypes carrying multiple disease resistance genes. The codominant Ty3-P6-25 marker was used to detect the Ty-3 gene associated with TYLCV resistance, whereas the C2-25 CAPS marker was employed to identify the Frl gene conferring resistance to FORL. Among the 167 breeding lines analyzed with the Ty3-P6-25 marker, 13 (9.7%) were homozygous resistant, 5 (3.7%) were heterozygous resistant, and 116 (86.6%) were homozygous susceptible, while 33 samples produced no interpretable results. Analysis of 177 breeding lines with the C2-25 marker identified 3 (2.5%) homozygous resistant, 10 (8.2%) heterozygous resistant, and 109 (89.3%) homozygous susceptible genotypes, whereas 55 samples yielded no evaluable results. The molecular analyses confirmed the presence of resistance alleles to both TYLCV and FORL within the breeding population and enabled the identification of elite breeding lines carrying resistance to multiple economically important pathogens when integrated with previous marker data for Mi-1 and Sw-5. These findings demonstrate the effectiveness of marker-assisted selection for the rapid identification of disease resistance genes and provide valuable genetic resources for pyramiding multiple resistance genes, accelerating tomato breeding, and supporting the development of sustainable, disease-resistant tomato cultivars.
