Investigation of in vitro Anti-trichomoniasis Effect of Nigella Sativa Oil against Trichomonas vaginalis
Abstract
Objective: Although 5-nitroimidazole derivatives (metronidazole, ornidazole) are being used in the current treatment of trichomoniasis, it has been known the presence of MTZ-resistant T. vaginalis strains for almost 50 years. Therefore, alternative treatment methods are being investigated for a long time for treatment of trichomoniasis. In this study, it was aimed to investigate in vitro activity of Nigella sativa oil against Trichomonas vaginalis. Material and Methods: In vitro testing of Nigella sativa and metronidazole against T. vaginalis was performed in cell culture plate by dilution method. Briefly, T. vaginalis trophozoites were transferred to 24-well plate containing TYM medium. Thereafter, different concentrations of Nigella sativa and metronidazole were added to each well and incubated at 37°C. Findings: At 48th hour of incubation, although motile trophozoites at 250 μg/ml concentration of Nigella sativa oil were observed, immotile trophozoites were detected at 500 μg/ml, 1000 μg/ml and 2000 μl/ml concentrations of Nigella sativa oil. At 72th hour, immotile trophozoites as well as trophozoite lysis were detected in all concentration of Nigella sativa oil, except for 250 μg/ ml concentration. Also at the end of 72th hour for Nigella sativa, MIC was 500 μl/mg while MLK was 1000μl/
mg. Conclusion: In this study, Nigella sativa oil was tested in vitro as an alternative therapeutic agent for treatment of trichomoniasis and it has been found that Nigella sativa oil has a naturally important therapeutic potential for trichomoniasis treatment. In vivo tests and the isolation and identification of compounds responsible for the detected activity are planned for further investigations
