Linking Total Cell Protein Profiles Revealed by SDS-PAGE in Erwinia amylovora Strains from Pear Orchards in Türkiye to Virulence Level Variation

Öz

Erwinia amylovora, the causal agent of fire blight, is a globally significant and highly destructive pathogen of pome fruit species, imposing severe economic constraints on sustainable agricultural systems. In this study, the total cellular protein profiles of 78 E. amylovora strains collected from pear orchards across seven distinct regions of Türkiye were analyzed to elucidate phenotypic relationships and assess intra-population diversity. Protein extraction was conducted following standardized protocols, and high-resolution SDS-PAGE was employed to generate reproducible electrophoretic fingerprints. The resulting banding patterns were quantitatively analyzed using GelJ v2 software, and phylogenetic relationships were inferred through cluster analysis. The SDS-PAGE-derived profiles revealed a predominantly conserved proteomic architecture, with overall similarity values ranging from 70% to 100%. Cluster analysis resolved the strains into five principal groups; however, a single dominant cluster encompassed the majority of isolates, indicating a largely homogeneous population structure. Despite this high degree of similarity, subtle yet consistent variations in band intensity and electrophoretic mobility were observed among selected strains, reflecting limited but biologically meaningful phenotypic heterogeneity. Notably, strain-specific deviations in the 35–55 kDa protein region and variations in band intensity, particularly among low-virulence isolates, suggest differential protein expression or possible post-translational modifications rather than large-scale structural proteomic divergence. Importantly, virulence levels exhibited a broad range (0–100%) but did not show a consistent correlation with overall protein profile similarity, indicating that pathogenic variability is likely governed by fine-scale regulatory mechanisms rather than gross proteomic differences. Geographical origin had no discernible impact on clustering patterns, as strains from diverse regions, were interspersed across the dendrogram. This finding suggests extensive epidemiological connectivity and a stable, widely distributed population structure across Türkiye.
