Sıcaklık Şoku Uygulaması ile Triploid Karadeniz Alabalığı (Salmo trutta labrax) Üretimi ve Kuluçka Performansının Belirlenmesi[*]

Öz

Bu çalışmada, triploid Karadeniz alabalığı (Salmo trutta labrax) üretmek için termal şok protokolünün optimizasyonu belirlenmiştir. Triploidizasyon işlemi için döllenmeden sonra 10, 15 ve 20. dakikalarda dört farklı sıcaklıkta (26, 28, 30 ve 32 ºC) şok uygulanmıştır. Ploidi oranı NOR (Nükleolar Organizatör Bölgeleri) boyama ve eritrosit büyüklüğü yöntemleri ile belirlenmiştir. En yüksek triploid oranları, döllendikten 15 dakika sonra 10 dakika sıcaklık şoku uygulamasıyla 32 ºC (%86,15) ve 28 ºC (%81,26)’lerde tespit edilmiştir. Fakat, çıkış oranı ve kuluçka randımanı 28 ºC sıcaklık şoku grubunda daha başarılı bulunmuştur. Çalışmadan elde edilen bulgular, Karadeniz alabalığı triploid uygulamalarında kullanılabilir.

Triploid Black Sea Trout (Salmo trutta labrax) Production by Using Heat Shocks and Determination of Incubation Performance

Öz

In this study, optimization of thermal shock protocol for triploid Black Sea trout (Salmo trutta labrax) production were determined. In total, four different temperature regimes (26, 28, 30, and 32 ºC) were performed following fertilization at 10th, 15 th, and 20 th minutes for triploidization process. Ploidy rate was determined by NOR (Nucleolus Organizer Regions) and measures of erythrocyte size. The highest rate of triploidy was detected with a shock treatment at 32 °C (86,15%) and 28°C (81,26%) for 10 min shock duration initiated 15 min post-fertilization. However, hatching rate and hatching efficiency were more successful at the 28 ºC heat shock group. The results of the present study is applicable to the production of triploid Black sea trout production.
