Cytological Evaluation of Vaginal Smear Samples in Rats

In order to increase the validity and reliability of the research results in some preclinical studies the estrus cycle phase of the experimental animal should be known in advance. Rats’ estrus cycle lasting as short as 4-5 days provides many advantages to the researchers and therefore it is important to assign the cycle period determinations. In rats with a regular estrus cycle, as a result of morphological changes occurring in the vaginal epithelium along with endocrine change there are cell types specific to each stage. In our study, it was aimed to introduce detailed cytological findings of rat estrus cycle periods. In this regard, from each of 28 Sprague Dawley female rat, vaginal smear samples were regularly taken and spread on slides. After the vaginal smear preparations were stained with Giemsa stain which gives rapid result, cytological imaging and identification of the cells were performed under the light microscope. The rat estrus cycles were determined with an interpret of small and large nuclei epithelial cells, keratinized epithelial cells without nuclei, and neutrophil leukocytes displayed in prepared preparations. After staining the cells of estrus cycle periods using Giemsa in rats, these cells can be evaluated cytological under the microscope easily and in a short time.

Sıçanlarda Vajinal Yayma Örneklerinin Sitolojik Değerlendirilmesi

Deney hayvanının östrüs siklus evresi bazı klinik öncesi çalışmalarda gerek araştırma sonuçlarının geçerliliğini gerekse de güvenilirliğini artırmak amacıyla önceden bilinmelidir. Sıçanların östrüs siklus döngülerinin 4-5 gün gibi kısa sürmesi araştırmacılara birçok yönden avantaj sağlar ve bu nedenle siklus dönem tayinlerinin belirlenmesini önemli kılar. Düzenli östrüs siklusu gösteren sıçanlarda, endokrin değişimle birlikte vajinal epitelde meydana gelen morfolojik değişiklikler sonucu her bir evreye özgü hücre tipleri bulunmaktadır. Bu çalışmada, sıçan östrüs siklus dönemlerinin ayrıntılı sitolojik bulgularının ortaya konulması amaçlandı. Çalışmamızda, 28 adet Sprague Dawley türü dişi sıçanların her birinden düzenli aralıklarla vajinal sürüntü örnekleri alındı ve lamlara yayıldı. Vajinal yayma preparatları hızlı sonuç veren Giemsa boyası ile boyandıktan sonra ışık mikroskobu altında hücrelerin sitolojik görüntülenmesi ve tanımlanması yapıldı. Hazırlanan preparatlarda görüntülenen çekirdekli küçük ve büyük epitelyal hücreler, çekirdeği olmayan keratinize epitelyal hücreler ve nötrofil lökositlerin yorumlanmasıyla sıçan östrüs siklus dönemleri belirlendi. Sıçanlarda östrüs siklusu dönemlerine ait hücrelerin Giemsa kullanılarak boyanmalarının ardından bu hücreler sitolojik olarak mikroskop altında kolaylıkla ve kısa bir süre içerisinde değerlendirilebilir.
