Antioxidant activity and tyrosinase enzyme inhibition potential of the endemic Stachys gaziantepensis and Stachys ketenoglui (Lamiaceae) from Turkey

Abstract

This study investigates the phytochemical profile and biological activities of two endemic Turkish species, Stachys gaziantepensis and Stachys ketenoglui. Aerial parts of the plants were extracted using ethyl acetate, methanol, and water through an ultrasound-assisted extraction process. The resulting extracts were evaluated in terms of total phenolic and flavonoid contents, antioxidant capacity (DPPH, ABTS, and metal chelation assays), and tyrosinase inhibitory activity. Among the tested samples, the ethyl acetate extract of S. gaziantepensis exhibited the highest phenolic and flavonoid levels. Methanol extracts of both species demonstrated superior radical scavenging performance in DPPH and ABTS assays, whereas the strongest metal-chelating effect was observed in the ethyl acetate extract of S. ketenoglui. Notably, the ethyl acetate extract of S. gaziantepensis showed pronounced tyrosinase inhibition. Overall, the findings indicate that both species possess considerable bioactive potential, with S. gaziantepensis displaying comparatively stronger activity. The results also emphasize the influence of solvent polarity on the extraction of biologically active compounds, suggesting that these endemic taxa may serve as promising candidates for pharmaceutical and cosmetic applications.
